三黄洗剂改善特应性皮炎小鼠模型皮肤屏障的机制探讨 被引量:1

摘要 目的探讨三黄洗剂改善特应性皮炎(AD)小鼠模型皮肤屏障可能的作用机制。方法取60只小鼠随机分为对照组、模型组、复方地塞米松软膏组、三黄洗剂低剂量组、中剂量组、高剂量组,涂抹2,4-二硝基氯苯丙酮溶液造模。每天涂抹给药2周后,采用免疫组化法检测中间丝聚合蛋白(FLG)、水通道蛋白3(AQP3)和角蛋白17(K17)的表达,称量皮肤湿重、干重,计算失重比。结果模型组、三黄洗剂低、中、高剂量组以及复方地塞米松乳膏组皮肤失重比比正常对照组增加且差异有统计学意义(P<0.01)。模型组FLG的表达明显低于正常对照组,三黄洗剂低、中、高剂量组和复方地塞米松乳膏组FLG表达量高于模型组。而模型组AQP3、K17的表达均明显高于正常对照组,三黄洗剂低、中、高剂量组和复方地塞米松乳膏组AQP3、K17表达量均低于模型组。结论三黄洗剂治疗AD小鼠后能减轻炎性水肿,提高FLG含量,降低AQP3、K17的表达,可证明其能促进皮肤屏障的修复。 Objective In order to explore the potential mechanism of Sanhuang lotion in improving skin barrier in atopic dermatitis(AD)mice models.Methods Sixty mice were randomly divided into control group,model group,compound dexamethasone ointment group,Sanhuang lotion low dose group,middle dose group and high dose group,and 2,4-dinitrochlorobenzene was applied.Two weeks after administration,the expression of filaggrin(FLG),aquaporin 3(AQP3)and keratin 17(K17)was detected by immunohistochemistry.The wet weight and dry weight of the skin were weighed,and the weight loss ratio was calculated.Results The skin weight loss ratio of the model group,Sanhuang lotion low,medium and high dose group and compound dexamethasone cream group was higher than that of the normal control group,and the differences were statistically significant(P<0.01).The expression of FLG in the model group was significantly lower than that in the normal control group.The expression of FLG in the low,medium and high dose groups of Sanhuang lotion and the compound dexamethasone cream group was higher than that in the model group.The expressions of AQP3 and K17 in the model group were significantly higher than those in the normal control group.The expression levels of AQP3 and K17 in the low,middle and high dose groups of Sanhuang lotion and compound dexamethasone cream group were lower than those in the model group.Conclusion Sanhuang lotion can reduce inflammation and edema,increase FLG and decrease the expression of AQP3 and K17 and repair the skin barrier.
