液相质谱联用法同时测定复方甘草片中甘草酸和吗啡

Simultaneous Determination of Glycyrrhizic Acid and Morphine in Compound Licorice Tablets by UPLC-MS/MS

目的建立同时测定复方甘草片中甘草酸和吗啡的含量测定方法。方法利用超高效液相色谱-串联质谱仪,采集方式多反应监测(MRM),Agilent Zorbax Hilic Plus C18色谱柱(2.1 mm×100 mm,1.8μm),以乙腈-15 mmol/L乙酸铵(50:50,v/v)为流动相进行洗脱,流速为0.25 ml/min,柱温40℃。结果甘草酸在1.91~19.1μg/ml范围内线性良好(r=0.9983),平均回收率96.5%,RSD=1.86%,吗啡在0.11~1.1μg/ml范围内线性良好(r=0.9994),平均回收率105.0%,RSD=1.90%。结论该方法灵敏度高,前处理简单,可用于复方甘草片的质量控制。

Objective To establish a method for the simultaneous determination of glycyrrhizic acid and morphine in Compound Licorice Tablets.Methods Ultra performance liquid chromatography-tandem mass spectrometer(UPLC-MS/MS)with multiple reaction monitoring(MRM)was used.An Agilent Zorbax Hilic Plus C_(18) column(2.1 mm×100 mm,1.8μm)was adopted.The mobile phase was acetonitrile-15 mmol/L ammonium acetate(50:50,v/v)with the flow rate of 0.25 ml/min,and the column temperature was 40℃.Results Glycyrrhizic acid had a good linearity in the range of 1.91-19.1μg/ml(r=0.9983)and the average recovery rate was 96.5%,with RSD of 1.86%.Morphine had a good linearity in the range of 0.11-1.1μg/ml(r=0.9994)and the average recovery rate was 105.0%,with RSD of 1.90%.Conclusion With the advantages of high sensitivity and simple pretreatment,this method can be used for the quality control of Compound Licorice Tablets.
