摘要
目的:探讨星形胶质细胞通过分泌IL-1β激活NF-κB通路促进脑胶质瘤生长的作用机制。方法:将星形胶质细胞和脑胶质瘤共培养,ELISA和Q-PCR分别检测星形胶质细胞中IL-1β的分泌和表达;CCK-8、Transwell及流式分别检测脑胶质瘤增殖、迁移和凋亡,蛋白免疫印迹检测NF-κB通路相关蛋白的表达。结果:CCK-8、Tran⁃swell及流式结果分别显示星形胶质细胞可以促进脑胶质瘤增殖、迁移和抗凋亡,而抑制星形胶质细胞中IL-1β的表达能抑制脑胶质瘤增殖、迁移和抗凋亡;蛋白免疫印迹结果显示抑制星形胶质细胞中IL-1β的表达能抑制脑胶质瘤中NF-κB通路的激活。结论:星形胶质细胞通过分泌IL-1β激活NF-κB通路促进脑胶质瘤增殖、迁移和抗凋亡的能力从而促进脑胶质瘤的生长,此项研究为揭示脑胶质瘤的恶性进展机制提供新的思路。
Objective To explore the mechanism by which astrocytes secrete IL-1βto activate the NF-κB pathway to pro⁃mote the growth of glioma.Methods Astrocytes and gliomas were co-cultured,and ELISA and Q-PCR were used to detect the secretion and expression of IL-1βin astrocytes.CCK-8,Transwell and flow cytometry were used to detect the prolifera⁃tion,migration and apoptosis of glioma cells.Western blotting was used to detect the expression of NF-κB pathway related proteins.Results CCK-8,transwell and flow cytometry showed that astrocytes could promote the proliferation,migration and anti-apoptosis of glioma,but inhibiting the expression of IL-1βin astrocytes could inhibit the proliferation,migration and anti-apoptosis of glioma.Western blot results showed that inhibiting the expression of IL-1βin astrocytes can inhibit the activation of NF-κB pathway in gliomas.Conclusion Astrocytes can promote the proliferation,migration and anti-ap⁃optosis of glioma by secreting IL-1βthereby promoting the growth of glioma.This study provides a new idea to reveal the mechanism of malignant progression of glioma.
