摘要
目的:探讨树鼩自发乳腺癌发生过程中的代谢变化,研究机体代谢物质的改变与肿瘤的发生、发展过程密切相关。方法:使用气相色谱—飞行时间质谱联用技术(GC-TOFMS)对血清代谢物进行非靶向测定,鉴定标注检测得到的代谢物,并进行代谢组学分析。结果:血清代谢组学发现14种代谢物浓度发生显著改变,与对照组相比,乳腺癌组中核糖酸,γ-内酯代谢物浓度显著上升,脯氨酸、苏氨酸、1,5-脱水葡萄糖醇、丝氨酸、花生四烯酸、肌酸等13种代谢物浓度显著下降。京都基因与基因组百科全书(KEGG)通路分析发现,花生四烯酸代谢、精氨酸和脯氨酸代谢这两个代谢通路发生显著改变。结论:自发乳腺癌树鼩血清中脯氨酸、肌酸、花生四烯酸等代谢物含量与正常树鼩相比有显著差异,通路分析提示精氨酸和脯氨酸代谢及花生四烯酸代谢途径极有可能影响乳腺癌的发生发展。
Objective:To explore the metabolic changes of spontaneous breast cancer in tree shrew,and to study whether the changes in the body's metabolites are closely related to the occurrence and development of tumors.Methods:Gas chromatography tandem time-of-flight mass spectrometry(GC-TOFMS)was used to perform nontargeted determination of serum metabolites,identify and label the detected metabolites,and perform metabonomics analysis.Results:Serum metabonomics revealed significant changes in the concentrations of 14 metabolites.Compared with the control group,the concentrations of ribonic acid,gamma-lactone metabolites in breast cancer group were significantly increased,while the concentrations of proline,threonine 1,1,5-anhydroglucitol,serine 1,arachidonic acid and creatine decreased significantly.Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis showed that arachidonic acid metabolism,arginine and proline metabolism were significantly changed.Conclusion:There were significant differences in serum proline,creatine,arachidonic acid and other metabolites in spontaneous breast cancer tree shrews compared with normal tree shrews.Pathway analysis suggested that arginine,proline and arachidonic acid metabolism may affect the occurrence and development of breast cancer.
