摘要
目的:探讨miR-372对结肠癌SW620细胞增殖、侵袭和迁移的影响及其机制。方法:将体外培养的SW620细胞分为对照组(未处理)、miR-NC组(转染miR-372模拟物阴性对照)、miR-372组(转染miR-372模拟物)、pLV-LATS2组(转染pLVLATS2质粒)和miR-372+LATS2组(转染miR-372模拟物和pLV-LATS2质粒),采用实时荧光定量PCR(qPCR)测定miR-372和LATS2 mRNA的表达,荧光素酶实验测定miR-372和LATS2的靶向关系,Western blotting测定LATS2蛋白的表达,MTT法测定各组细胞的增殖,Transwell小室实验测定细胞的侵袭和迁移。结果:与对照组相比,miR-372组细胞中miR-372表达水平升高,而LATS2 mRNA表达降低(P<0.05);LATS2是miR-372的潜在靶基因。与对照组相比,miR-372组细胞中LATS2蛋白的表达水平显著降低,细胞的增殖水平显著升高、侵袭和迁移细胞数显著增多,而pLV-LATS2组中LATS2蛋白的表达水平显著升高,细胞的增殖水平显著降低、侵袭和迁移细胞数显著减少,且miR-372+LATS2组细胞增殖水平、侵袭细胞数和迁移细胞数均显著低于miR-372组(P<0.05)。结论:miR-372可靶向LATS2促进结肠癌SW620细胞增殖、侵袭和迁移。
Objective:To investigate the effect and mechanism of miR-372 on the proliferation,invasion and migration of colon cancer SW620 cells.Methods:SW620 cells were cultured in vitro and transfected with miR-NC,miR-372 mimic,or pLV-LATS2 plasmid,and co-transfected with miR-372 mimic and pLV-LATS2 plasmid.The cells in the control group were not transfected.The expressions of miR-372 and LATS2 mRNA were measured by qPCR.The targeting relationship between miR-372 and LATS2 was determined by luciferase assay.The expression of LATS2 protein was determined by Western blotting.The cell proliferation was measured by MTT assay,and cell invasion and migration abilities were detected by Transwell.Results:Compared with the control group,the expressions of miR-372,the proliferative capacity and the number of invaded and migrated cells were increased,while the mRNA and protein expression of LATS2 was decreased in cells transfected with miR-372 mimic(P<0.05).LATS2 was a target gene of miR-372.The expression level of LATS2 protein was increased significantly,while the proliferative capacity and the number of invaded and migrated cells were evidently reduced in cells transfected with pLV-LATS2 plasmid(P<0.05).Compared with miR-372 group,the proliferation,invasion and migration abilities were lower in cells co-transfected with miR-372 mimic and pLV-LATS2(P<0.05).Conclusion:miR-372 promotes the proliferation,invasion and migration of colon cancer SW620 cells by targeting LATS2.
