RBM25在结直肠癌中的表达及其对细胞增殖的影响 被引量:1

The expression of RBM25 in colorectal cancer and its effect on cell proliferation

摘要 目的探究RNA结合基序蛋白25(RBM25)在结直肠癌组织中的表达及对结直肠癌细胞功能的影响。方法利用TCGA数据库分析RBM25在结直肠癌和正常结肠组织中的表达差异及RBM25表达水平与患者预后之间的关系;分别采用qRT-PCR和Western blot检测结直肠癌组织中RBM25在mRNA和蛋白水平的表达情况。将RBM25-siRNAs转染结直肠癌细胞系HCT 116,采用CCK-8实验、克隆形成实验检测RBM25敲低对结直肠癌细胞生长、增殖的影响;利用TCGA数据库分析RBM25 mRNA表达水平与结直肠癌患者预后之间的相关性。结果对TCGA数据库分析显示,RBM25 mRNA在结直肠癌与正常组织的表达差异无统计学意义(P>0.05),但RBM25 mRNA水平与患者的预后呈负相关(P<0.05);qRT-PCR结果显示RBM25 mRNA在结直肠癌患者癌组织中高表达,但差异无统计学意义(P>0.05),Western blot结果显示RBM25蛋白在结直肠癌组织中高表达(P<0.05);RBM25-siRNAs敲低导致HCT 116细胞生长、增殖能力下降(P<0.05)。结论RBM25蛋白在结直肠癌组织中高表达,并促进结肠癌细胞的生长、增殖,提示RBM25可能与结直肠癌的发生、发展相关。 Objective To investigate the expression of RNA binding motif protein 25(RBM25)in colorectal cancer(CRC)tissues and further explore its effects on the function of CRC cells.Methods The TCGA database was used to analyze the differential expression of RBM25 in CRC and corresponding normal tissues,and the correlation between the expression of RBM25 mRNA and the prognosis of CRC patients.QRT-PCR and Western blot were conducted to determine the expression of RBM25 in the mRNA and protein levels in clinical CRC tissues and paired normal tissues,respectively.CRC cell line HCT 116 was transfected with RBM25-siRNAs,and then CCK-8 experiments and colony formation experiments were performed to investigate the effects of RBM25 knockdown on the growth and proliferation of CRC cells.Results TCGA database analysis showed that the differential expression of RBM25 mRNA between CRC tissues and corresponding normal tissues was not statistically significant(P>0.05),but the level of RBM25 mRNA was negatively correlated with the patient′s prognosis(P<0.05);The results of qRT-PCR showed that RBM25 mRNA was higher in CRC tissues than that in paired normal tissues,but the differential expression was not statistically significant(P>0.05).Western blot results showed that RBM25 was upregulated in CRC tissues(P<0.05);RBM25-siRNA knockdown retarded the growth and proliferation of CRC cell line HCT 116(P<0.05).Conclusion The upregulation of RBM25 protein promotes the growth and proliferation of CRC cells,suggesting that RBM25 may be related to the occurrence and progression of CRC.
