TRPV1、CRF1在反流性食管炎大鼠下丘脑和食管组织中的表达 被引量:1

Expression of TRPV1 and CRF1 in hypothalamus and esophagus in rats with reflux esophagitis

摘要 目的分析反流性食管炎(reflux esophagitis,RE)大鼠模型的下丘脑和食管组织中TRPV1、CRF1的表达,初步探讨TRPV1、CRF1在RE发病机制中的作用。方法采用贲门肌部分切开+幽门半结扎法构建9只RE大鼠模型,以9只假手术大鼠作为对照组。应用免疫组化、Western blotting、qRT-PCR分别检测RE大鼠和对照组大鼠下丘脑、食管下段组织中TRPV1、CRF1的蛋白、mRNA表达情况。结果RE大鼠模型食管大体标本呈充血表现,但未见明显糜烂、溃疡,下丘脑组织大体标本无明显变化。RE大鼠下丘脑TRPV1、CRF1的蛋白表达显著高于对照组大鼠、mRNA相对表达量显著高于对照组大鼠。RE大鼠和对照组大鼠下丘脑组织TRPV1蛋白表达秩均值、秩和分别是13.44 vs 5.56、120 vs 50;RE大鼠和对照组大鼠下丘脑组织CRF1蛋白表达秩均值、秩和分别是12.78 vs 6.22、115 vs 56。RE大鼠与对照组大鼠食管下段组织TRPV1、CRF1蛋白、mRNA表达比较,差异均无统计学意义(P>0.05)。结论RE大鼠下丘脑组织中TRPV1、CRF1表达显著升高,其可能在中枢层面参与RE的病理过程,这为进一步研究RE的发病分子机制提供新的视角。 Objective To analyze the expression of TRPV1 and CRF1 in hypothalamus and esophagus tissues of reflux esophagitis(RE)rat models,and to explore the role of TRPV1 and CRF1 in the pathogenesis of RE.Methods Partial incision of cardia muscle and semi-ligation of pylorus were used to construct RE rat models,sham-operated rats were used as the control group.Immunohistochemistry,Western blotting and qRT-PCR were used to detect the expression of TRPV1 and CRF1 protein in the hypothalamus and lower esophagus tissues of RE rats and control rats.Results The general esophagus specimens of RE rats showed hyperemia,but no erosions and ulcers.There was no obvious change in the general hypothalamic tissue specimens.The protein expression of TRPV1 and CRF1 in the hypothalamus of RE rats was significantly higher than that of control group rats,and the relative mRNA expression was significantly higher than that of control group rats.The average rank and rank sum of TRPV1 protein expression in RE rats and control group rats were 13.44 vs 5.56,121 vs 50,respectively.The average rank and rank sum of CRF1 protein expression in RE rats and control group rats were 12.78 vs 6.22,115 vs 56,respectively.There was no significant difference in the expression of TRPV1 and CRF1 in the lower esophageal tissue between RE rats and control group rats(P>0.05).Conclusion TRPV1 and CRF1 in the hypothalamus tissue of RE rats are significantly increased,which may participate in the pathological process of RE at the central level,it provides a new perspective for further research on the molecular mechanism of RE pathogenesis.
