摘要
目的:比较银屑病患者皮损与健康人皮肤间充质干细胞(MSCs)生物学特性的差异。方法:分离和培养银屑病组(15例)与健康对照组(15例)皮肤MSCs,比较二者生长速度、基因表达差异及细胞因子分泌水平。结果:与健康对照组相比,银屑病组皮损MSCs的生长速度明显缓慢(P<0.05)。2组基因测序结果示共鉴定出1539个差异表达的mRNA,其中130个在银屑病组皮损MSCs中表达上调,1409个表达下调,且JAK-STAT信号通路上有17个基因在银屑病组皮损MSCs中表达下调。2组细胞因子分泌水平比较,在增殖相关的细胞因子中,银屑病组表皮生长因子(EGF)和干细胞因子(SCF)表达均明显高于健康对照组(P<0.05),而碱性成纤维细胞生长因子(bFGF)表达明显减少(P<0.05);在炎症相关细胞因子中,银屑病组白细胞介素(IL)-11浓度显著高于健康对照组(P<0.05),而IL-3、IL-6、IL-8和肝细胞生长因子(HGF)浓度均明显低于健康对照组(P<0.05)。结论:银屑病患者皮损MSCs生物学特性有异常,可能是导致患者皮损局部微环境异常的原因及银屑病的发病机制之一。
Objective:To compare the biological characteristics of skin mesenchymal stem cells(MSCs)between psoriasis patients and controls.Methods:The specimens were divided into psoriasis group and healthy control group with 15 cases in each group.MSCs were isolated from the skin lesions of patients and healthy controls,and then the growth rate,gene expression differences and the levels of cytokine secretion of the two groups were compared.Results:Compared with healthy control group,the growth rate of MSCs in psoriasis group was significantly lower(P<0.05).A total of 1539 differentially expressed m RNAs were identified by sequencing,of which 130 were up-regulated and 1409 were down-regulated in psoriasis group.Notably,17 genes in the JAKSTAT signaling pathway were down-regulated in psoriasis group.The expression of EGF and SCF in the psoriasis group were significantly higher than those in healthy control group(P<0.05),while the expression of bFGF was significantly reduced(P<0.05).The concentration of IL-11 in the psoriasis group was significantly higher than that in healthy control group(P<0.05),while the IL-3,IL-6,IL-8 and HGF were significantly lower than those in healthy control group(P<0.05).Conclusion:The abnormal biology of mesenchymal stem cells may disrupt the microenvironment to trigger the pathogenesis of psoriasis.
