摘要
目的探究P53信号通路在苦参碱诱导人结肠癌细胞凋亡的作用机制;方法通过使用苦参碱干预人结肠癌SW-480细胞,使用CCK-8、Western blot实验检测细胞P53基因以及侵袭力的变化;结果 CCK-8实验,苦参碱干预结肠癌细胞,使其增殖力降低(P<0.05);Western blot实验,苦参碱组较对照组P53/Bax蛋白表达量升高(P<0.05),E-Cad蛋白表达量升高(P<0.05)。结论苦参碱可通过P53信号通路促进人结肠癌SW-480细胞凋亡,从而诱导细胞凋亡,降低结肠癌细胞的侵袭和转移能力,达到抗肿瘤作用。
Objective To explore the mechanism of P53 signaling pathway in matrine induced apoptosis of human colon cancer cells;Methods Use matrine to intervene in human colon cancer SW-480 cells, and use CCK-8 and Western blot to detect P53 gene and Changes in invasiveness;Results CCK-8 experiment, matrine intervenes in colon cancer cells to reduce their proliferative power P<0.05;Western blot experiment, matrine group increased expression of P53/Bax protein compared with control group High P<0.05,E-Cad protein expression increased P<0.05.Conclusion Matrine can promote the apoptosis of human colon cancer SW-480 cells through P53 signaling pathway, thereby inducing apoptosis, reducing the invasion and metastasis ability of colon cancer cells, and achieving anti-tumor effect.
