CALML3鉴别诊断肺腺癌和鳞状细胞癌的潜在价值分析

Analysis of the potential value of CALML3 in the differential diagnosis of lung adenocarcinoma and squamous cell carcinoma

目的探讨钙调蛋白样蛋白3(CALML3)鉴别诊断肺鳞状细胞癌(LUAD)和肺腺癌(LUSC)的潜在价值。方法采用生物信息学的方法分析CALML3在LUAD和LUSC中的表达情况;分析LUAD、LUSC临床特征对CALML3表达水平的影响;采用荧光定量PCR(qPCR)检测CALML3 mRNA在LUAD和LUSC中的表达情况;采用受试者工作特征(ROC)曲线分析CALML3对LUAD、LUSC的诊断价值。结果通过对癌症基因组图谱(TCGA)数据分析发现CALML3在LUSC中表达水平明显升高,在LUSC中的改变倍数是LUAD中的7149倍。LUSC患者性别、年龄、吸烟情况、肿瘤分期及淋巴结转移程度对CALML3的相对表达水平没有影响(P>0.05)。CALML3在LUSC、LUAD组织中的相对表达水平高于癌旁正常组织(P<0.05)。CALML3在LUSC中的曲线下面积(AUC)为0.837。结论CALML3可作为区分LUAD和LUSC的候选标志物。

Objective To explore the potential value of calmodulin-like 3(CALML3)in the differential diagnosis of lung squamous cell carcinoma(LUAD)and lung adenocarcinoma(LUSC).Methods Bioinformatics methods were used to analyze the expression of CALML3 in LUAD and LUSC;the influence of LUAD and LUSC clinical characteristics on the expression of CALML3 was analyzed;fluorescent quantitative PCR(qPCR)was used to detect the expression of CALML3 mRNA in LUAD and LUSC.The receiver operating characteristic(ROC)curve was used to analyze the diagnostic value of CALML3 for LUAD and LUSC.Results Through the analysis of the cancer genome atlas(TCGA)data,it was found that the expression level of CALML3 in LUSC was significantly increased,and the fold change in LUSC was 7149 times that in LUAD.The gender,age,smoking status,tumor stage and lymph node metastasis of LUSC patients had no effect on the relative expression level of CALML3(P>0.05).The relative expression level of CALML3 in LUSC and LUAD tissue was higher than that in the adjacent normal tissue(P<0.05).The area under the curve(AUC)of CALML3 in LUSC was 0.837.Conclusion CALML3 can be used as a candidate marker to distinguish LUAD from LUSC.
