应用Illumina MiSeq测序技术对腹泻北平顶猴肠道菌群的分析 被引量:3

Gut Microbiome Analysis of Diarrheal Northern Pig-Tailed Macaque(Macaca leonina)Based on High-Throughput Sequencing Technology

摘要 为探寻可能导致北平顶猴(Macaca leonina)腹泻的菌群及其变化规律,分别采集6只健康和6只腹泻北平顶猴的新鲜粪便,应用Illumina MiSeq高通量测序技术测定其肠道菌群的结构以及腹泻过程中肠道菌群的动态变化。结果表明:健康组肠道菌群的Shannon指数(P=0.015)、Chao 1指数(P=0.014)、ACE指数(P=0.032)较腹泻组高。在门水平上,腹泻组肠道中拟杆菌门(Bacteroidetes)、变形菌门(Proteobacteria)和未鉴定细菌(unidentified Bacteria)的平均相对丰度明显高于健康组,而厚壁菌门(Firmicutes)平均相对丰度低于健康组。在属水平上,腹泻组肠道中的乳杆菌属(Lactobacillus)、普氏菌属(Prevotella)和未鉴定瘤胃球菌属(unidentified Ruminococcaceae)的平均相对丰度明显低于健康组,而链球菌属(Streptococcus)、粪杆菌属(Faecalibacterium)、弯曲杆菌属(Campylobacter)的平均相对丰度明显高于健康组。腹泻与健康北平顶猴肠道菌群丰度具有显著的差异,揭示了这些差异菌群可能是北平顶猴腹泻的重要因素之一。 In order to study the gut flora causing diarrhea of northern pig-tailed macaque(Macaca leonina)and their patterns,the fresh feces were collected from six healthy and diarrheic adult northern pig-tailed macaque,the composition and function of their gut flora were analyzed.Alpha diversity index analysis showed that the Shannon index(P=0.015),Chao1 index(P=0.014)and ACE index(P=0.032)of gut flora were significantly higher in healthy group than those in diarrheic group.At the phylum level,relative abundance of Bacteroidetes,Proteobacteria and unidentified Bacteria in diarrheic group were higher than that in healthy group,relative abundance of Firmicutes was lower than that in healthy group.At the genus level,relative abundance of Lactobacillus,Prevotella and unidentified Ruminococcaceae in diarrhea group were lower than that in healthy group,relative abundance of Streptococcus,Faecalibacterium,and Campylobacter were higher than that in healthy group.The significant difference in microbial abundance between healthy and diarrheic northern pig-tailed macaque suggests that some microbial communities may be one of the important factors causing diarrhea.
