摘要
为探究治疗驴沙门菌病的新方法,以驴源马流产沙门菌为宿主菌,利用双层平板法进行沙门菌烈性噬菌体的分离鉴定及生物学特性研究。结果分离到1株长尾噬菌体vB-SabS-Sds2,对3株驴源马流产沙门菌裂解率为100%,对17株鸡白痢沙门菌裂解率为6%。最佳感染复数为0.000 1,噬菌体效价达3.23×109 PFU/mL。该噬菌体潜伏期为10 min,爆发期为30 min,爆发量为8.6 PFU/cell,对氯仿不敏感,对紫外线较为敏感,在高温及pH 3~13环境中均有较高活性。结果表明噬菌体vB-SabS-Sds2作为1株烈性噬菌体对驴源马流产沙门菌具有广泛的裂解能力,具有较强的特异性和抵抗力。本试验为驴沙门菌病的噬菌体治疗提供了科学依据。
To explore a new method for the treatment of donkey abortion induced by Salmonella abortus equi, lytic bacteriophage was isolated using S. abortus equi from donkey as host bacteria and its biological characteristics were determined by double agar plate method. The resulting phage strain named vB-SabS-Sds2 belonging to Siphovirus showed cleavage efficiency for 3 S. abortus equi strains and 17 Salmonella pullorum strains at 100% and 6%, respectively. When the MOI of bacteriophage was 0.000 1, the phage titer could reach 3.23×109 PFU/mL. The phage demonstrated the incubation period of 10 min, the outbreak period of 30 min, and the burst size of 8.6 PFU/cell. Being insensitive to chloroform, but sensitive to ultraviolet, the bacteriophage was resistant to temperature and active at pH 3 to 13. Considering its high specificity and extensive lytic ability for S. abortus equi, phage vB-SabS-Sds2 offers a potential bacteriophage treatment for donkey salmonellosis.
