蛹虫草多糖对微波辐射损伤雄性小鼠生殖功能的影响 被引量:1

Effect of Cordyceps Militaris Polysaccharide on Male Reproductive Function in Microwave Radiation Injury Mice

摘要 为了观察蛹虫草多糖(CMP)对微波辐射损伤雄性小鼠生殖功能的影响,并探讨其初步机制。将75只清洁级雄性昆明小鼠随机分成对照组、模型组、蛹虫草多糖低、中、高剂量组,除对照组外,将其余各组小鼠置于10 mW/cm2微波辐射环境1 h/d,连续30 d。最后1次照射后6 h进行小鼠性行为能力测试。微波辐射后,蛹虫草多糖低、中、高剂量组灌胃给药[0.3 mL/(10 g·bw)]蛹虫草多糖10 mg/(10 g·bw)、20 mg/(10 g·bw)、40 mg/(10 g·bw),连续30 d,最后1次灌胃后6 h进行小鼠性行为能力测试,12 h后取材,进行精子相对计数、精子畸形率、精子死亡率检测;血清中氧化损伤指标、睾酮(TTE)含量检测。结果显示:与对照组比较,模型组小鼠的扑捉潜伏期(CIP)显著延长、扑捉次数(CT)显著减少(P<0.05);精子相对计数、精子存活率显著减少(P<0.05);精子畸形率显著增加(P<0.05);血清中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)的活性以及TTE水平显著降低(P<0.05);血清中丙二醛(MDA)的含量显著增加(P<0.05)。与模型组比较,蛹虫草多糖高剂量组CIP显著缩短、CT显著增加(P<0.05);精子相对计数、精子存活率显著增加(P<0.05);精子畸形率显著减少(P<0.05);血清中SOD、GSH-Px的活性以及TTE水平显著升高(P<0.05);血清中MDA的含量显著降低(P<0.05)。结果表明蛹虫草多糖能够保护微波辐射损伤雄性小鼠的生殖功能,其作用机制可能与减轻生殖细胞的氧化损伤有关。 In order to investigate if cordyceps militaris polysaccharide improves male reproductive function in microwave radiation injury mice and its potential mechanism, 75 male Kunming mice of clean grade were randomly divided into normal control group, microwave radiation group, low-, medium-dose and high-dose of cordyceps militaris polysaccharide groups(15 mice per group). Except for normal control group, other groups were exposed to 10 mW/cm2 microwave radiation for 1 h/d in 30 consecutive days. Sexual behavior of mice was tested 6 h after the last irradiation. After microwave irradiation, the low-, middle-dose and high-dose groups were administered orally of cordyceps militaris polysaccharides \[10 mg/(10 g·bw), 20 mg/(10 g·bw) and 40 mg/(10 g·bw), respectively\] for 30 consecutive days, and sexual behavior of mice was tested 6 h after the last gastric administration. Sperm relative count, sperm deformity rate, sperm survival rate, the index of oxidative damage and the level of testosterone(TTE) in sera were also measured 12 h later. After microwave radiation, capture incubation period(CIP) of mice was prolonged, the capture time(CT) was reduced(P<0.05), relative sperm count, the activity of the superoxide dismutase(SOD),glutathione peroxidase(GSH-Px) and the level of TTE in sera and sperm survival rate decreased(P<0.05), while sperm deformity rate and the content of malondialdehyde(MDA) in sera increased(P<0.05). It was of note that cordyceps militaris polysaccharide high-dose group significantly shortened the capture latency, increased the capture time(P<0.05), sperm relative count, sperm survival rate(P<0.05), and SOD, GSH-Px activity and TTE level(P<0.05), but decreased sperm deformity rate(P<0.05), and the content of MDA in serum(P<0.05). The results indicated that cordyceps militaris polysaccharide can affect male reproductive system, and its mechanism may be related to the oxidative damage of germ cells.
