上游刺激因子1对脑胶质瘤微血管内皮细胞增殖、迁移及体外血管生成的影响

摘要 目的探究上游刺激因子1(Upstream stimulating factors 1,USF1)对脑胶质瘤微血管内皮细胞(Glioma microvascular endothelial cell,GEC)的增殖、迁移及体外血管生成等方面的影响。方法构建GEC模型以及USF1过表达和表达沉默的稳定转染细胞系,Western blot用于检测USF1蛋白的表达,CCK-8法用于检测细胞增殖能力,体外血管生成试验用于检测血管生成能力,Transwell细胞迁移试验用于检测细胞迁移能力。结果上调USF1表达可以增强GEC的增殖、迁移和血管生成能力,而下调USF1表达则降低GEC的增殖、迁移和血管生成能力。结论USF1过表达对GEC的增殖、迁移和血管生成能力等生物学行为具有增强作用,抑制USF1的表达能够降低GEC增殖、迁移和血管生成能力。 Objective To investigate the effect of upstream stimulating factors 1(USF1)on proliferation,migration and tube formation of glioma microvascular endothelial cell(GEC).Methods Established GEC model and stable transfected cell lines of overexpression and knockdown of USF1.Western blot was used to detect the expression of USF1 protein.CCK-8 test was used to detect cell proliferation,tube formation assay was used to evaluate the tube formation,and Transwell cell migration test was used to detect cell migration.Results Upregulation of USF1 expression enhanced the proliferation,migration and angiogenesis of GEC,while downregulation of USF1 expression decreased the proliferation,migration and angiogenesis of GEC.Conclusion Overexpression of USF1 in GEC can promote proliferation,migration and tube formation.Suppression of USF1 expression can reduce proliferation,migration and tube formation.
