卡介苗促进肺间充质干细胞IL-10和TGF-β1表达

BCG promotes the expression of IL-10 and TGF-β1 in lung-resident mesenchymal stem cells

目的研究卡介苗(Bacillus Calmette-Guérin, BCG)对肺间充质干细胞(lung-resident mesenchymal stem cells, LR-MSCs)增殖、细胞凋亡、细胞因子及相关蛋白表达,探讨LR-MSCs在结核病(tuberculosis, TB)发生、发展中的作用。方法采用CCK-8法检测细胞增殖、流式细胞术检测细胞凋亡及表面蛋白(CD45、CD19、CD34、CD14、CD90、CD105、CD29、CD44和CD73)、ELISA检测IL-10含量、Western Blot检测相关蛋白表达(STAT3、p-STAT3和TGF-β1)。结果 BCG对LR-MSCs细胞增殖(P=0.530,P>0.05)、凋亡(P=0.650,P>0.05)、表面标志物表达和相关蛋白STAT3表达均无显著影响(P>0.05);BCG促进LR-MSCs细胞IL-10产生(P<0.05)、TGF-β1和p-STAT3表达(P<0.05)。结论 BCG感染可能通过促进LR-MSCs细胞IL-10和TGF-β1表达,激活STAT3磷酸化,从而影响TB的发生、发展。

Objective To study the effects of Bacillus Calmette-Guérin(BCG) on the proliferation, apoptosis, cytokine, and related protein expression of lung-resident mesenchymal stem cells(LR-MSCs) and explore the role of LR-MSCs in tuberculosis(TB). Methods CCK-8 test was used to detect cell proliferation, flow cytometry was used to detect LR-MSCs’ apoptosis and surface proteins(CD45, CD19, CD34, CD14, CD90, CD105, CD29, CD44 and CD73), besides, detection of IL-10 concentration by ELISA and expression of related proteins(STAT3, p-STAT3 and TGF-β1) by Western Blot. Results BCG had no significant effect on the proliferation(P=0.530,P>0.05), apoptosis(P=0.650,P>0.05) and surface proteins(P>0.05) of LR-MSCs;BCG promoted the production of IL-10 in LR-MSCs cells(P<0.05), and induced expression of TGF-β1 and p-STAT3(P<0.05;P<0.05), while had no effect on STAT3 protein expression(P>0.05). Conclusion BCG infection may mediate the occurrence and development of TB by promoting the expression of IL-10 and TGF-β1 in LR-MSCs and activating STAT3 phosphorylation.
