骨碎补对东莨菪碱模型小鼠学习记忆能力的影响 被引量:1

摘要 目的探讨骨碎补改善东莨菪碱模型小鼠学习记忆能力的作用机制。方法昆明小鼠分为空白组、东莨菪碱模型组、骨碎补组、盐酸多奈哌齐组、抗脑衰胶囊组、ER阻断剂组。行为学测试小鼠学习记忆能力;HE染色观察小鼠海马形态;免疫组化法检测小鼠海马中雌激素受体ERα、ERβ、FSHR、LHR表达;ELISA测定小鼠海马中ACh含量、AChE、ChAT活性。结果与模型组对比,行为学测试各给药组小鼠学习记忆能力明显改善(P<0.01);海马神经元结构清晰、排列紧密;ERα、ERβ、FSHR、LHR表达增加(P<0.05或P<0.01);ACh含量增加(P<0.01)、ChAT的活性增加(P<0.01)、AChE活性减弱(P<0.01),且均被ER阻断剂逆转。结论ER通路介导骨碎补改善东莨菪碱模型小鼠胆碱能系统,增强学习记忆能力。 Objective To explore the effect of Drynaria fortunei on improving the learning and memory abilities of scopolamine model mice.Methods The experiment is divided into control group,scopolamine model group,Drynaria fortunei group,donepezil hydrochloride group,anti-brain failure capsule group,ER antagonist group.Behavioral test of learning and memory ability of mice;HE staining to observe the morphology of mouse hippocampus;Immunohistochemical method to detect the expression of ERα,ERβ,FSHR and LHR in the hippocampus of mice;ELISA is used to determine the ACh content,AChE and ChAT activity in the hippocampus of mice.Results Compared with the model group,the learning and memory abilities of mice in each administration group are significantly improved in the behavioral test(P<0.01);the hippocampal neurons are clearly structured and arranged closely;Increased expression of ERα,ERβ,FSHR and LHR(P<0.05 or P<0.01);ACh content increased(P<0.01),ChAT activity increased(P<0.01),AChE activity decreased(P<0.01).And they are all reversed by ER blockers.Conclusion The ER pathway mediates Drynaria fortunei to improve the cholinergic system of scopolamine model mice and enhance the ability of learning and memory.
