人参皂苷Rg1对高糖诱导的肾小管上皮细胞损伤保护作用 被引量:6

摘要 目的探讨人参皂苷Rg1对高糖诱导的人肾小管上皮细胞HK-2中炎症反应和纤维化的影响及可能的机制。方法细胞计数法(CCK-8)检测不同浓度人参皂苷Rg1对细胞活性的影响。HK-2细胞随机分为正常对照组、高糖组、低、高人参皂苷Rg1组。ELISA检测TNF-α和IL-1β,Western blot分析α-SMA、E-cadherin、Fn、TGF-β1、Smad 2和Smad 3的表达情况。结果人参皂苷Rg1对HK-2细胞无明显毒性作用。与正常对照组相比,高糖组TNF-α和IL-1β含量升高(P<0.05),α-SMA、Fn、TGF-β1、Smad 2和Smad 3的表达水平上调(P<0.05),E-cadherin的表达水平下调(P<0.05);与高糖组相比,低、高人参皂苷Rg1组TNF-α和IL-1β含量降低(P<0.05),α-SMA、Fn、TGF-β1、Smad 2和Smad 3的表达水平下调(P<0.05),E-cadherin的表达水平上调(P<0.05)。结论人参皂苷Rg1通过抑制HG诱导的肾小管上皮细胞炎症反应和EMT来阻碍肾纤维化进展,其机制可能与阻断TGF-β/Smad信号通路有关。 Objective To investigate the effect of ginsenoside Rg1 on inflammation and fibrosis in human renal tubular epithelial cells HK-2 induced by high glucose and its possible mechanism.Methods Cell counting method(CCK-8)is used to detect the effect of different concentrations of ginsenoside Rg1 on cell viability.HK-2 cells are randomly divided into normal control group,high glucose group,low and high ginsenoside Rg1 groups.ELISA detects TNF-αand IL-1β,western blot analysis ofα-SMA,E-cadherin,Fn,TGF-β1,Smad 2 and Smad 3 expression.Results Ginsenoside Rg1 has no obvious toxic effect on HK-2 cells.Compared with the normal control group,the levels of TNF-αand IL-1βin the high glucose group are increased(P<0.05),and the expression levels ofα-SMA,Fn,TGF-β1,Smad 2 and Smad 3 are up-regulated(P<0.05),E-cadherin expression level is down-regulated(P<0.05);compared with the high glucose group,the levels of TNF-αand IL-1βin the low and high ginsenoside Rg1 groups are reduced(P<0.05),the expression level ofα-SMA,Fn,TGF-β1,Smad 2 and Smad 3 are down-regulated(P<0.05),and the expression level of E-cadherin is upregulated(P<0.05).Conclusion Ginsenoside Rg1 inhibits the progression of renal fibrosis by inhibiting HGinduced inflammatory response and EMT in renal tubular epithelial cells.The mechanism may be related to blocking the TGF-β/Smad signaling pathway.
