摘要
陆生植物具有特殊的解毒和修复功能,通过酶促代谢反应降低或清除体内的异源有机毒素物质。为筛选和鉴定拟南芥解毒修复相关的调控因子,以对毒素超敏感的ROXY19转基因植株为亲本,通过正向遗传学手段筛选抗性恢复的突变体。初筛和复筛获得8个解毒能力相对提高的候选突变体,命名为biànhuó1~8(bió1~8)。与野生型拟南芥和亲本植株相比,bió1突变体主根毛数量明显增加,bió2突变体花瓣数目变多。bió候选突变体呈现不同生长发育表型,说明植物的解毒修复反应和生长发育过程紧密相关,可能发现新的解毒修复调控因子。
Green plants in natural environments respond rapidly to the presence of xenobiotic substances and detoxify them into less harmful chemicals through phytoremediation.We attempted to develop a versatile and reliable forward genetic screening system to isolate xenobiotic detoxification genes in model plant Arabidopsis.We screened for xenobiotic resistant mutants from EMS-mutagenized ROXY19 ectopic expression plants which were hyper-sensitive to xenobiotic stresses.In total,we found eight xenobiotic resistant mutants and named them biànhuó1–8(bió1–8).The mutant candidate bió1 displayed increased root hairs and bió2 showed more flower petals than wild-type and mother plants.Our results indicate that plant detoxification response and development process are tightly regulated,and we may find novel regulation factors of the xenobiotic detoxification pathway that await further investigations.
