杨树ERF、MYB和NAC转录因子家族基因耐盐胁迫表达特性分析 被引量:5

Expression characteristics analysis of ERF,MYB and NAC transcription factor family genes in poplar under salt stress

摘要 植物转录因子是一种广泛存在于植物体中,参与植物生长发育调控和抗逆胁迫过程的重要基因家族。为筛选杨树耐盐胁迫相关转录因子,分析其分子生物学功能及应答盐胁迫表达模式,本研究采用生物信息学方法,筛选出杨树ERF、MYB、NAC三类转录因子家族中与盐胁迫相关的目的基因20个。对其理化性质、序列同源性、启动子顺式作用元件及基因功能注释进行系统分析,发现有2个基因(Potri.012G060300和Potri.017G082500)参与了刺激响应过程。同时,基于RNA-seq测序数据表达模式分析发现,在正常条件下Potri.012G060300基因在芽、叶、茎组织中的表达量较高;盐胁迫处理后,表达量均降低。Potri.017G082500基因在正常条件下芽中的表达量较高,在茎中的表达量相对较低;在盐胁迫处理后,该基因在芽和叶中的表达量均较高,在茎中的表达量较低。此外,通过对20个基因进行表达相关性分析,发现3组基因(7个)间存在较强的表达相关性。 Plant transcription factors(TFs)are widely distributed in plant and involved in the regulation of plant growth and development and stress resistance.In order to screen TFs related to salt stress tolerance in poplar and analyze their molecular biological functions and expression patterns in response to salt stress,20 target genes related to salt stress in ERF,MYB and NAC transcription factor families of poplar were selected using bioinformatics.The physicochemical properties,sequence homology,promoter cis-acting elements and gene function annotation were systematically analyzed.Two genes(Potri.012G0060300 and Potri.017G082500)were found to be involved in the salt response process.The expression pattern analysis based on RNA-seq data showed that the expression level of Potri.012G0060300 in buds,leaves and stems were higher under normal conditions,while the expression levels were decreased under salt stress treatment.The expression level of Potri.017G082500 was higher in buds and lower in stems under normal conditions,and under salt stress treatment,the expression level of this gene was higher in buds and leaves,but lower in stems.In addition,expression of genes in the 3 groups showed strong correlation.
