过表达LINC00595降低前列腺癌细胞PC3对多西他赛的耐药性 被引量:1

摘要 目的探讨LINC00595对前列腺癌细胞PC3对多西他赛(DTX)耐药性的影响。方法以DTX为诱导药物构建人前列腺癌耐药细胞株PC3-DTX,计算IC50值;RT-qPCR检测LINC00595及酪氨酸激酶受体(Trk)B在PC3细胞系和PC3-DTX耐药细胞系中的表达水平;MTS实验、流式细胞和克隆形成实验分别分析过表达LINC00595对PC3-DTX细胞增殖、周期和凋亡及细胞生长的影响。结果PC3-DTX细胞组对DTX IC50值显著高于正常培养PC3细胞组(P<0.001)。PC3-DTX细胞中LINC00595相对表达水平显著低于PC3细胞(P<0.01),而PC3-DTX细胞中TrkB mRNA相对表达水平显著高于PC3细胞。与ov-NC组相比,过表达LINC00595显著抑制PC3-DTX细胞增殖和克隆形成,促进细胞周期从S期向G2期转化、细胞凋亡,降低PC3-DTX细胞中TrkB mRNA相对表达。结论过表达LINC00595降低前列腺癌细胞PC3对DTX的耐药性。 Objective To investigate effects of LINC00595 on docetaxel(DTX)resistance of prostate cancer PC3 cells.Methods The human prostate cancer drug-resistant cell line PC3-DTX was constructed with DTX as the inducing drug,and its IC50 value was calculated.The expression levels of LINC00595 and Tyrosine kinase receptor B(TrkB)in PC3 cells and PC3-DTX cells were measured using RT-qPCR.The effects of LINC00595 over-expression on the proliferation,cell cycle,apoptosis and cell growth of PC3-DTX cells were assessed using MTS assay,flow cytometry and colony formation assay,respectively.Results The IC50 value of PC3-DTX cells to DTX was significantly higher than that of normal PC3 cells(P<0.001).The relative expression level of LINC00595 in PC3-DTX cells was significantly lower than that in PC3 cells(P<0.01),while the relative expression level of TrkB mRNA in PC3-DTX cells was significantly higher than that in PC3 cells.Compared with the control group,over-expression of LINC00595 significantly inhibited the proliferation and clone formation,promoted the cell cycle transformation from S phase to G2 phase and apoptosis,and decreased TrkB mRNA expression in PC3-DTX cells.Conclusions Over-expression of LINC00595 reduces DTX resistance in prostate cancer PC3 cells.
