抑制miR-128-3p对阿尔茨海默症大鼠认知功能障碍的影响 被引量:4

摘要 目的探讨抑制miR-128-3p对阿尔茨海默症(AD)大鼠认知功能障碍的影响。方法24只大鼠随机分为模型组、假手术组、miR-128-3p antagomir组、antagomir NC组,通过RT-qPCR检测各组大鼠海马组织miR-128-3p表达情况,水迷宫实验观察各组大鼠学习记忆能力,尼氏染色法观察各组大鼠海马组织细胞形态结构变化,TUNEL检测各组大鼠海马组织细胞凋亡情况,Western-blot检测大鼠海马组织NR2B、GluR1、Bcl-2、Bax表达水平。结果与假手术组比,模型组大鼠海马组织miR-128-3p高表达(P<0.05),逃避潜伏期显著延长,平台停留时间显著减少(P<0.05),CA1和CA3区神经元和尼氏小体数量显著下降,细胞着色较浅,萎缩呈空泡样,细胞凋亡率显著升高,海马组织NR2B、GluR1、Bcl-2表达水平显著下降,Bax表达水平显著升高(P<0.05)。与antagomir NC组比,miR-128-3p antagomir组大鼠海马组织miR-128-3p表达水平显著下降(P<0.05),逃避潜伏期显著缩短,平台停留时间显著延长(P<0.05),CA1和CA3区神经元和尼氏小体数量较多,细胞圆润饱满,凋亡率显著下降,海马组织NR2B、GluR1、Bcl-2表达水平显著升高,Bax表达水平显著下降(P<0.05)。结论抑制miR-128-3p能够改善AD大鼠学习和记忆能力,并抑制海马神经元细胞凋亡。 Objective To investigate the effect of inhibition miR-128-3 p on cognitive dysfunction in Alzheimer’s disease(AD)rats.Methods 24 rats were randomly divided into model group,control group,miR-128-3 p antagomir,antagomir NC group,each rat hippocampus by RT-qPCR detection miR-128-3 p expression,water maze experiment observation group rats learning and memory ability,Nissl staining to observe each group rats hippocampal tissue cell morphology change,TUNEL detect each rat hippocampus tissue apoptosis,Western blot detection rat hippocampal tissue NR2 B,GluR1,Bcl-2,Bax expression levels.Results Than with the control group,model group rats hippocampus miR-128-3 P high expression(P<0.05),to escape significantly prolong the incubation period,platform residence time was significantly reduced(P<0.05),CA1 and CA3 area of neurons and a significant reduction in the number of nissl bodies,cell coloring shallow,contraction cavity sample,cell apoptosis rate increased significantly,hippocampus NR2 B,GluR1,Bcl-2 expression level decreased significantly,Bax expression level increased significantly(P<0.05).Than to the antagomir NC group,miR-128-3 P antagomir group rat hippocampus miR-128-3 P expression level decreased significantly(P<0.05),the escape latency was significantly shortened,platform for significantly prolonged(P<0.05),the CA1 and CA3 area neurons and nissl body number,rounded cells full of apoptosis rate dropped significantly,hippocampus NR2 B,GluR1,a significant rise in the Bcl-2 expression level,Bax expression level decreased significantly(P<0.05).Conclusion Inhibition of Mir-128-3 P can improve the learning and memory ability of AD rats and inhibit the apoptosis of hippocampal neurons.
