基于肠肾轴理论探讨三七注射液对慢性肾脏病大鼠肠黏膜功能和肠道菌群的影响 被引量:11

摘要 目的探讨三七注射液对慢性肾衰竭大鼠肠黏膜和肠道菌群的影响。方法将120只SD大鼠随机分为6组:正常组、模型组、三七注射液低剂量组、三七注射液中剂量组、三七注射液高剂量组、阳性对照组。用生化检测、ELISA、鲎试剂检测和细菌平板计数法分别检测肾功能、硫酸吲哚酚、血清D-乳酸、血浆内毒素、回肠内容物细菌培养鉴定及肠道菌群移位情况。结果 ①模型组大鼠较正常组大鼠肌酐、尿素氮、硫酸吲哚酚、D-乳酸、内毒素、肠道细菌移位率显著升高,致病菌大肠杆菌、肠球菌明显增多,益生菌双歧杆菌、乳杆菌明显减少。②与模型组比较,三七中剂量组、三七高剂量组、阳性对照组大鼠肌酐、尿素氮、硫酸吲哚酚、D-乳酸、内毒素、肠道细菌移位率显著降低,且致病菌大肠杆菌、肠球菌明显减少,益生菌双歧杆菌、乳杆菌明显增多。结论慢性肾功能衰竭会使大鼠肠道菌群失调,代谢毒素增多,肠黏膜屏障功能损伤,肠通透性增加,肠道菌群发生移位;三七注射液可以改善上述情况,以三七中、高剂量组作用显著,提示三七注射液延缓CKD大鼠肾纤维化进展的机制可能与其调节CKD大鼠肠道菌群,修复肠道黏膜屏障功能及抑制肠道菌群移位有关。 Objective To investigate the effects of Sanqi Injection(三七注射液) on intestinal mucosa and intestinal flora in rats with chronic renal failure. Methods A total of 120 SD rats were randomly divided into 6 groups: normal group, model group, low dose group of Sanqi Injection, medium dose group of Sanqi Injection, high dose group of Sanqi Injection and positive control group. Renal function, indoxol sulfate, serum D-lactic acid, plasma endotoxin, ileum contents, bacterial culture and intestinal flora translocation were detected by biochemical assay, ELISA, limulus reagent and bacterial plate count. Results Compared with those of the normal group, the levels of creatinine, urea nitrogen, indolephenol sulfate, D-lactic acid and endotoxin and intestinal bacterial translocation rate in the model group were significantly increased. The escherichia coli and enterococcus of the model group were significantly increased, and the probiotics bifidobacterium and lactobacillus of the model group were significantly decreased. Compared with those of the model group, the levels of creatinine, urea nitrogen, indolephenol sulfate, D-lactic acid and endotoxin and intestinal bacterial translocation rate in rats of Sanqi Injection medium and high dose groups and positive control group were significantly reduced. The escherichia coli and enterococcus were significantly decreased, and the probiotics bifidobacterium and lactobacillus were significantly increased. Conclusion Chronic renal failure can cause intestinal flora imbalance, metabolic toxin increase, intestinal mucosal barrier function damage, intestinal permeability increase and intestinal flora shift. Sanqi Injection can improve the above situation, with significant effect in medium and high dose groups of Sanqi Injection, suggesting that the mechanism of Sanqi Injection in delaying the progression of renal fibrosis in chronic renal failure rats may be related to the regulation of intestinal flora in chronic renal failure rats, repair of intestinal mucosal barrier function and inhibition of intestinal flora translocation.
