摘要
采用70%乙腈溶液提取花生、玉米样品,然后用免疫磁珠固相萃取、净化和富集提取液中的黄曲霉毒素B_(1),高效液相色谱荧光检测仪定量分析。对影响免疫磁固相萃取的条件包括提取剂种类、吸附剂用量、吸附时间、洗脱液类型、洗脱液体积进行优化。结果表明,黄曲霉毒素B_1在0.5~50μg/kg范围内线性关系良好,相关系数为0.999 5,回收率为90.3%~115.7%,相对标准差小于6.9%,检出限和定量限分别为0.1μg/kg和0.3μg/kg。方法步骤简单、准确性好、检出限低,能够应用于粮油作物产品中花生、玉米黄曲霉毒素B_1的定量检测。
The peanut and corn samples were extracted with 70%acetonitrile solution,and purified and enriched by immunomagnetic solid-phase extraction beads(IMSPE).Aflatoxin B_(1)(AFB_(1))was determined by high performance liquid chromatography with fluorescence detector(HPLC-FLD).The IMSPE conditions,including extractant,adsorbent amount,adsorption time,the type and volume of eluent were studied.Under the optimized conditions,a good linear relationship was established in the range of 0.5~50μg/kg and the correlation coefficient was 0.9995.The recovery was ranging from 90.3%to 115.7%and RSDs less than 6.9%.The detection limit(LOD)and the quantification limit(LOQ)were 0.1μg/kg and 0.3μg/kg,respectively.The IMSPE-HPLC-FLD method was simple,fast,high accuracy and with low detection limit,which could be applied for the determination of AFB 1 inagri-products.
