摘要 目的建立苍术超高效液相色谱(UPLC)指纹图谱,并同时测定苍术中绿原酸和苍术素的含量,为苍术的质量控制提供依据。方法建立15批苍术药材样品UPLC指纹图谱,同时测定苍术素、绿原酸的含量,通过相似度计算结合聚类分析(HCA)、主成分分析(PCA)及正交偏最小二乘-判别分析(OPLS-DA)对15批样品进行质量分析。结果苍术UPLC指纹图谱共标识出25个共有峰,经鉴定,其中2个共有峰为绿原酸和苍术素。15批苍术样品指纹图谱相似度为0.738~0.997;HCA和PCA将15批样品分为三类,OPLS-DA共标记和筛选出7个差异标志物;山东样品中苍术素含量最高、绿原酸含量最低,内蒙古和河北样品中2种成分含量相差不大。结论本研究建立的指纹图谱和多成分定量方法稳定可靠,可为苍术的质量控制提供参考。 Objective To establish the UPLC fingerprint of Atractylodis Rhizoma and determine the contents of chlorogenic acid and atractylodin simultaneously;To provide references for the quality control of Atractylodis Rhizoma.Methods The UPLC fingerprint of 15 batches of Atractylodis Rhizoma was established.The contents of chlorogenic acid and atractylodin were determined simultaneously.The quality of 15 batches was analyzed by similarity calculation combined with HCA,PCA and OPLS-DA.Results A total of 25 common fingerprint peaks were identified,two of which were identified as chlorogenic acid and atractylodin.The fingerprint similarity of 15 batches of samples ranged from 0.738 to 0.997.HCA and PCA divided 15 batches of samples into 3 groups.Seven differential markers were identified by OPLS-DA.The content of atractylodin in samples from Shandong Province was the highest and the content of chlorogenic acid was the lowest.There was no significant difference between samples from Inner Mongolia and Hebei.Conclusion The established fingerprint and multi-components quantitative method are stable and reliable,which can provide reference for the quality control of Atractylodis Rhizoma.
