摘要
目的:优化胡黄连苷的提取工艺。方法:以出膏率、胡黄连苷Ⅰ和胡黄连苷Ⅱ转移率为考察指标,以提取溶剂种类(水、乙醇)、液料比、提取次数和提取时间为考察因素,在单因素试验基础上,运用Box-Behnken响应面法进行优化,确定了提取胡黄连苷的最佳工艺条件,并进行实验验证。结果:优化的提取工艺为95%乙醇提取3次,液料比20∶1,提取时间为4.5 h。结论:Box-Behnken响应面优化的提取工艺可用于胡黄连苷的提取。
Objective:To optimize the extraction process of picroside from Picrorhizae Rhizoma.Methods:With extraction rate and transfer rates of picrosideⅠand picrosideⅡas evaluation indexes,types and volume fractions of extraction solvents(water,ethanol),liquid-to-material ratio,extraction times,and extraction duration were investigated through single factor test and Box-Behnken design for the purpose of optimizing the extraction process of picroside from Picrorhizae Rhizoma.The resultant extraction parameters were verified.Results:The optimal extraction parameters are as below:liquid-to-material ratio of 20,extraction solvent of 95%ethanol,extraction for 4.5 h,with a total of 3 times.Conclusion:Box-Behnken can be used for optimizing the extraction process of picroside from Picrorhizae Rhizoma.
