摘要
目的:建立西洋参破壁粉碎前后高效液相色谱法(HPLC)指纹图谱,并采用双标线性校正法进行定性。方法:采用高效液相色谱-电雾式检测器法(HPLC-CAD),以西洋参破壁饮片与传统饮片为研究对象,使用相似度分析进行质量评价。另以人参皂苷Re和人参皂苷Rb2为双标化合物,使用DRSOrigin软件进行双标线性校正,预测特征峰的保留时间,并比较其与相对保留时间法的差别,以辅助色谱峰定性分析。结果:所选10批西洋参破壁饮片与传统饮片指纹图谱相似度均在0.939以上,共确定17个共有峰,并建立了以人参皂苷Rg_(1)、Re、Rb_(1)、Rc、Rb_(2)、Rb_(3)、Rd及拟人参皂苷F_(11)为特征峰的对照特征图谱。双标线性校正法预测保留时间的精度优于相对保留时间法。结论:所建立的方法简便、准确,可为该品种的质量评价与控制提供有力的技术支持。同时,可为其他破壁饮片质量标准研究提供参考。
Objective:To establish the high performance liquid chromatography(HPLC)fingerprints of Panacis Quinquefolii Radix Pieces before and after cell wall-breaking and conduct the qualitative analysis by linear calibration using two reference substances.Methods:The HPLC with charged aerosol detector(HPLC-CAD)was employed for similarity analysis and quality evaluation of cell wall-broken Panacis Quinquefolii Radix pieces and traditional pieces.In addition,ginsenosides Re and Rb_(2)were used as double standard compounds for linear calibration using DRS Origin,followed by the prediction of the retention time of characteristic peaks and its comparison with the retention time predicted by relative retention time method to assist the qualitative analysis of chromatographic peaks.Results:The fingerprint similarity between the 10 batches of cell wall-broken Panacis Quinquefolii Radix pieces and traditional pieces was all above 0.939,and 17 common peaks were identified.A reference chromatogram with ginsenosides Rg_(1),Re,Rb_(1),Rc,Rb_(2),Rb_(3),Rd and pseudo-ginsenoside F_(11)as characteristic peaks was established.The accuracy of retention time predicted by LCTRS was better than that by the relative retention time method.Conclusion:The method established in this study is simple and accurate,which has provided a technical support for the quality evaluation and control of cell wall-broken Panacis Quinquefolii Radix pieces and also some reference for the quality standard research of other cell wall-broken pieces.
