摘要
目的研究假酸浆果Nicandra physaloides(L.) Gaertn.的化学成分及其抗炎活性。方法假酸浆果70%乙醇提取物采用硅胶、ODS、Sephadex LH-20和HPLC进行分离纯化,根据理化性质及波谱数据鉴定所得化合物的结构。采用RAW264.7模型评价其抗炎活性。结果从中分离得到27个化合物,分别鉴定为咖啡酸甲酯(1)、巴拉诺酚B(2)、4-O-阿魏酸-奎尼酸甲酯(3)、异地黄苷(4)、7α-柑橘B(5)、7β-柑橘B(6)、金雀异黄素7-O-β-D-阿哌呋喃磺酰基-(1→6)-O-β-D-吡喃葡萄糖苷(7)、(+)-柚皮苷(8)、(-)-柚皮苷(9)、异槲皮素(10)、槲皮素3-O-(6″-O-α-鼠李糖-β-半乳吡喃糖苷)(11)、异荭草苷(12)、5,7,3′,4′-四羟基-8-C-β-D-葡萄糖黄酮碳苷(13)、小茴香素葡萄糖(14)、木黄酮(15)、1-甲基-1,2,3,4-四氢-β-咔啉-3-羧酸(16)、苦杏仁苷(17)、N-乙酰色氨酸甲酯(18)、沙棘酰胺(19)、canusesnol A(20)、麦芽糖(21)、3-羧乙基-3-羟基戊二酸1,5-二甲酯(22)、withahisolide G(23)、daturataturin A(24)、withastramonolide(25)、阿魏酰酪胺(26)、N-(E)-p-coumaroyltyramine(27)。化合物1、3~4、6~11、13~15、18、20、22~27均显示出对炎症细胞NO的生成起抑制作用。结论所有化合物均为首次从该植物中分离得到。化合物1、3~4、6~11、13~15、18、20、22~27具有潜在的抗炎活性。
AIM To study the chemical constituents from the fruits of Nicandra physaloides(L.) Gaertn.and their anti-inflammatory activities.METHODS The 70% ethanol extract from N. physaloides was isolated and purified by silica, ODS,Sephadex LH-20 and HPLC,then the structures of obtained compounds were identified by physicochemical properties and spectral data. Their anti-inflammatory activities were evaluated by RAW264.7 model.RESULTS Twenty-seven compounds were isolated and identified as caffeic acid methyl ester(1), balanophonin B(2), 4-O-ferulic-quinic acid methyl ester(3), isomartynoside(4), 7α-citrusin B(5), 7β-citrusin B(6), genistein7-O-β-D-apiofuranosyl-(1→6)-O-β-D-glucopyranoside(7),(+) narigin(8),(-) narigin(9), isoquercitrin(10), quercetin 3-O-(6″-O-α-rhamnopyranosyl-β-galactopyranoside)(11), isoorientin(12), 5,7,3′,4′-tetrahydroxy-8-C-β-D-glucopyranosylflavone(13), ononin glucoside(14), xylogranatinin(15), 1-methyl-1,2,3,4-tetrahydro-β-carboline-3-carboxylic acid(16), amygdalin(17), N-acetyltrypto-phan methyl ester(18), hippophamide(19), canusesnol A(20), maltose(21), 3-carboxyethyl-3-hydroxyglutaric acid 1,5-dimethyl ester(22), withahisolide G(23), daturataturin A(24), withastramonolide(25), feruloyltyramine(26), N-(E)-p-coumaroyltyramine(27). Compounds 1,3-4,6-11,13-15,18,20,22-27 showed inhibitory effects on NO production in inflammatory cells.CONCLUSION All the compounds are isolated from this plant for the first time. Compounds 1,3-4,6-11,13-15,18,20,22-27 may have potential anti-inflammatory activities.
