The Association between 5HT2A T102C Polymorphisms and Behavioral and Psychological Symptoms in Alzheimer’s Disease:A Meta-Analysis

目的:研究5-HT2A T102C基因多态性与阿尔茨海默病精神行为症状的相关性。方法:检索PubMed、Web of Science、Cochrane、中国知网和万方数据库,时间自建库至2020年9月。经文献筛选、质量评价、数据提取后,使用stata 15统计结果。结果:共纳入9项研究,2113位患者。结果显示5-HT2A T102C基因多态性与幻觉、淡漠和异常运动行为的发生有关:T等位基因变异至C后,幻觉发生的可能性增加(P=0.023)。TT基因型较非TT基因型(CC+TC)发生幻觉(P=0.000)和淡漠(P=0.035)的可能性减小。TC基因型较非TC基因型(CC+TT)发生幻觉(P=0.005)和异常运动行为(P=0.043)的可能性增加。5-HT2A T102C基因多态性在妄想、激越和抑郁上无统计学意义。结论:5-HT2A T102C基因多态性可能与阿尔茨海默病幻觉、淡漠和异常运动行为有关。

Objective:To review the association of 5-HT2 A T102 C polymorphisms with behavioral and psychological symptoms in Alzheimer’s Disease.Methods:Studies were collected using PubMed,Web of Science,the Cochrane Library,Chinese National Knowledge Infrastructure(CNKI),and Wanfang databases.After searching the literature,assessment of the literature’quality,and extraction data,we utilized the stata 15 software to analyze the results.Results:We included nine clinical studies with 2113 subjects.Our results showed that 5-HT2 A T102 C polymorphisms had an association with hallucination,apathy,and aberrant motor behavior symptoms.When the T allele mutates into the C allele,the presence of hallucination symptoms might be increased(P=0.023).TT genotype might be a protective factor when compared with nonTT genotype(CC+TC)which decreased the presence of hallucination(P=0.000)and apathy(P=0.035)symptoms.TC genotype might acted as an adverse factor when compared with non-TC genotype(CC+TT)which increased the presence of hallucination(P=0.000)and aberrant motor behavior(P=0.043)symptoms.No associations were observed between 5-HT2 A T102 C polymorphisms with delusion,agitation,and depression.Conclusion:5-HT2 A T102 C polymorphisms may have association with hallucination,apathy,and aberrant motor behavior symptoms in Alzheimer’s disease.
