摘要
目的探讨肝性脑病大鼠OXA神经元表达数量、活性变化及其临床意义。方法将雄性Wistar大鼠随机分为实验组和对照组,进行Morris水迷宫实验检测空间学习和记忆能力,Real-time qPCR方法检测前食欲素原(prepro-orexin)mRNA表达数量,免疫荧光法分别检测OXA、c-Fos表达。结果实验组血氨浓度显著高于对照组(P<0.05),空间学习和记忆能力较对照组显著降低(P<0.05),prepro-orexin mRNA的表达及OXA阳性神经元数量组间差异无统计学意义(P>0.05),但是实验组OXA/c-Fos双染阳性的神经元数量显著低于对照组(P<0.01)。结论肝性脑病大鼠空间学习及记忆能力降低,或许与激活状态的OXA神经元减少相关,而与OXA神经元总数量无关。
Objective To investigate the expression quantity of OXA neuron,the change of activity and its clinical significance in rats with hepatic encephalopathy.Methods Male Wistar rats were randomly divided into experimental group and control group.Morris Water Maze was used to measure their spatial learning and memory,and real-time qPCR was employed to detect mRNA expression of prepro-orexin,and the expression of OXA,while c-fos were detected by immunofluorescence assay.Results Compared with the control group,the serum ammonia level of the experimental group was significantly higher(P<0.05),and the spatial learning and memory abilities were obviously reduced(P<0.05).The expression of prepro-orexin mRNA and the number of OXA positive neurons showed no significant difference between these groups(P>0.05),but the number of OXA/c-Fos double-positive neurons in the experimental group was significantly lower than that in the control group(P<0.01).Conclusion The decrease of spatial learning and memory ability in rats with hepatic encephalopathy may be related to the decrease of activated OXA neurons,but not to the total number of OXA neurons.
