正交试验结合特征图谱法优选蛇蚁喷雾剂渗漉工艺 被引量:2

摘要 目的优选蛇蚁喷雾剂乙醇渗漉工艺。方法以芍药苷、丹酚酸B与延胡索乙素的综合保留率为评价指标,以乙醇体积分数、粉碎粒度、渗漉流速、溶剂用量为考察因素,采用L9(34)正交试验筛选渗漉工艺参数,并建立特征图谱,以相似度验证优选工艺的合理性。结果渗漉工艺影响因素大小依次为乙醇体积分数>溶剂用量>渗漉流速>粉碎粒度。渗漉最佳工艺条件为,赤芍、丹参、延胡索等18味药材粉碎成粗粉,加2倍量70%乙醇浸渍24 h,加10倍量70%乙醇进行渗漉,渗漉流速为9 mL/(min·kg)。与传统工艺相比,9个正交渗漉工艺12个共有峰的相似度均大于0.90。结论优选的工艺稳定、合理,可为蛇蚁喷雾剂的规模化生产提供理论依据。 Objective To optimize ethanol percolation process for Sheyi Sprays.Methods Taking the comprehensive retention rate of paeoniflorin,salvianolic acid B and tetrahydropalmatine B as the evaluation index and the volume fraction of ethanol,solvent dosage,crushing particle size and flow rate of percolation as the investigation indexes,the percolation process parameters were selected by the L9(34)orthogonal test,and the specific chromatogram was established to verify the rationality of the optimized process by the similarity.Results The influencing factors of percolation process were as follows:ethanol volume fraction>solvent dosage>solvent dosage>flow rate of percolation>crushing particle size.The optimal percolation process conditions were as follows:18 medicinal flavors such as Paeoniae Radix Rubra,Salvia Miltiorrhizae Radix et Rhizoma and Corydalis Rhizoma were crushed into the coarse powder,soaked in two times of 70%ethanol for 24 h,and percolated with 12 times of 70%ethanol with the flow rate of 9 mL/(min·kg).Compared with the traditional process,the similarity of 10 common peaks of nine orthogonal percolation processes was higher than 0.90.Conclusion The optimial process is stable and reasonable,which can provide a theoretical basis for the large-scale production of Sheyi Sprays.
