多指标-响应曲面法优选连翘叶中苷类有效部位的提取工艺

摘要 目的采用多指标-响应曲面法优化连翘叶中苷类有效部位的提取工艺。方法采用高效液相色谱法(high performance liquid chromatography,HPLC)检测连翘叶中有效成分含量,以连翘叶中的连翘酯苷A、芦丁、连翘苷的含量为评价指标,通过单因素及响应曲面法考察提取时间、乙醇体积分数、提取次数、料液比对连翘叶中苷类有效部位提取工艺的影响。结果最佳提取工艺:提取溶媒为40%乙醇,提取时间1.5 h,提取次数3次,料液比1∶14;此时综合评分值Y为98.06%。结论运用多指标-响应曲面法对连翘叶中苷类有效部位的提取工艺进行优化,该方法简便准确,易于操作,专属性强,可用于连翘叶中苷类有效部位的提取。 Objective To optimize the extraction process of the effective parts of glycosides in the leaves of Forsythia suspensa using multi-index-response surface method.Methods High performance liquid chromatography(HPLC)was used for content determination of active ingredients in Forsythia suspensa leaves.With the content of forsythiaside A,rutin and forsythin as the evaluation indexes,the effects of extraction time,ethanol volume fraction,extraction times,and solid-liquid ratio on the extraction process of the effective parts of glycosides were investigated by the single factor and response surface methodology.Results Optimal parameters were as follows:the extraction solvent of 40%ethanol,extraction time for 1.5 h,extraction for three times,and liquid to solid ratio of 1∶14.Under those conditions,the comprehensive score value Y was 98.06%.Conclusion The extraction process of glycosides from the leaves of Forsythia suspensa is successfully optimized by multi-index response surface method.This optimized extraction technology is simple,accurate,feasible,and has high specificity,and it could be used in the extraction of the effective parts of glycosides from the leaves of Forsythia suspensa.
