麦冬有效部位对支原体感染小鼠肺组织水通道蛋白1、5表达的影响 被引量:10

摘要 目的:观察麦冬有效部位对肺炎支原体感染小鼠肺组织水通道蛋白1(AQP 1)、水通道蛋白5(AQP 5)表达的影响,探讨其润肺生津作用。方法:将100只BALB/c小鼠,随机分为正常对照组,模型组,阿奇霉素组,养阴清肺组,麦冬多糖高、中、低剂量组和麦冬皂苷高、中、低剂量组,每组10只。除正常对照组外,其余各组采用MP滴鼻法造模,连续造模3 d后,各组小鼠开始灌胃给药,每天1次,阿奇霉素连续给药5 d,其他组连续给药6 d后,采集小鼠肺组织,光镜观察病理变化,RT-PCR测定肺组织AQP 1、AQP 5 mRNA的表达。结果:病理结果显示:与正常组比较,模型组可见明显炎症反应;与模型组比较,阿奇组、养阴清肺组、麦冬多糖高、中剂量组及麦冬皂苷低剂量组的肺组织炎症明显减轻,具有极显著差异。肺组织AQP 1 mRNA的表达:与正常对照组比较,模型组表达极显著下调(P<0.01);与模型组比较,麦冬皂苷低剂量组和麦冬多糖高、中、低剂量组的表达量提高,具有极显著性差异(P<0.01),阿奇霉素组及麦冬皂苷中剂量组表达量提高,具有显著性差异(P<0.05)。肺组织AQP 5 mRNA表达:与正常对照组比较,模型组表达极显著下调(P<0.01);与模型组比较,麦冬皂苷低剂量组和麦冬多糖中、低剂量组的表达量提高,具有极显著性差异(P<0.01),麦冬多糖高剂量组、阿奇霉素组及麦冬皂苷中剂量组表达量提高,具有显著性差异(P<0.05)。结论:麦冬有效部位(多糖、皂苷)均可上调支原体肺炎小鼠肺组织AQP 1、AQP 5表达,减轻肺组织病理变化;发挥润肺生津作用。 Objective:To investigate the effects of effective fractions of Ophiopogon on the expressions of aquaporin 1(AQP 1)and aquaporin 5(AQP 5)in the lung tissue of mice infected with Mycoplasma in order to explore the moistening lung and promoting fluid production function of Ophiopogon.Methods:100 BALB/c mice were divided into normal group,model group,Azithromycin group,Yangyin Qingfei group,Ophiopogon polysaccharide high,medium and low dose group,Ophioposide high,medium and low dose group,each group contained 10 mice.Except the normal group,the other groups rats were molded by MP nasal drip method.After continuous modeling for 3 days,oral administration was started.Azithromycin was administered continuously for 5 days,the other medicines were continuously administered for 6 days,once a day.Lung tissue was collected,pathological changes were observed by microscope,the expressions of AQP 1 and AQP 5 mRNA in lung tissue were determined by RT-PCR.Results:Pathological results showed that model group showed obvious inflammation(P<0.01)to compared with normal group;Compared with model group,lung inflammation in Azithromycin group,Yangyin Qingfei group,Ophiopogon polysaccharide high-dose,medium-dose and ophiopogon saponin low-dose groups was significantly reduced(P<0.01).AQP 1 mRNA:compared with the normal group,the AQP 1 mRNA expression in the lung tissue of mice in model group was significantly lower(P<0.01);compared with the model group,the expression of AQP 1 mRNA in Ophiopogon japonicus saponin low dose group and three dose of Ophiopogon japonicus polysaccharide groups was raised significantly(P<0.01),that in Azithromycin group and Ophiopogon japonicus saponin middle dose group was raise,too(P<0.05).AQP 5 mRNA:compared with the normal group,the AQP 5 mRNA expression in the lung tissue of mice in model group was significantly lower;compared with the model group,the expression of AQP 5 mRNA in Ophiopogon japonicus saponin low dose group and Ophiopogon japonicus polysaccharide middle,low dose groups was increased significantly(P<0.01),that in Ophiopogon japonicus saponin middle dose dose group,Ophiopogon japonicus polysaccharide high dose group and Azithromycin group was increased,too(P<0.05).Conclusion:Ophiopogon effective fractions can up-regulating AQP 1 and AQP 5 mRNA expressions in lung tissue of mice infected with Mycoplasma,reduce pathological changes,play a role in the function of moistening lung and promoting fluid production.
