千金藤素对胰腺癌细胞凋亡作用的研究

摘要 目的研究千金藤素(CEP)对胰腺癌细胞AsPC-1细胞凋亡的影响。方法配制不同浓度(0、2.1875、4.375、8.75、17.5、35μmol/L)的CEP作用于AsPC-1细胞,48h后通过MTT法检测CEP对AsPC-1细胞存活率的影响,镜下观察不同浓度药物作用后细胞生长状态变化,台盼蓝染色计算细胞存活率,Western blot检测凋亡相关蛋白聚腺苷酸二磷酸核糖基聚合酶(PARP)、剪切型胱天蛋白酶3(cleaved-caspase-3)及剪切型半胱氨酸的天冬氨酸蛋白水解酶9(cleaved-caspase-9)的表达量;流式细胞术检测细胞凋亡率。结果CEP可以明显抑制AsPC-1细胞的增殖(P<0.05);与正常对照组细胞相比,CEP作用后细胞死亡率增加(P<0.05);随着给药浓度的增加凋亡相关蛋白PARP、cleaved-caspase-3及cleaved-caspase-9的蛋白表达量明显增加;流式细胞凋亡结果显示CEP作用后AsPC-1细胞凋亡率均有所增高。结论CEP能够通过凋亡机制抑制胰腺癌细胞的增殖。 Objective To investigate the effect of Cepharanthine(CEP)on apoptosis of pancreatic cancer cell line AsPC-1.Methods Different concentration(0,2.1875,4.375,8.75,17.5,35μmol/L)of stephania action was first prepared and then exerted an effect on AsPC-1 cell.After 48hours,the MTT method was used to detect the effect of CEP on the survival rate of AsPC-1 cell,cell growth at was detected by the microscopic observation at different drug concentration,the cell survival rate was calculated by trypan blue staining.Western blot was conducted to detect the expression of apoptosis-related proteins PARP,cleaved-caspase-3 and cleaved-caspase-9,and apoptosis rate was detected by flow cytometry.Results CEP could significantly inhibit the proliferation of AsPC-1 cells(P<0.05);When compared with the negative control cells,the cell death rate increased(P<0.05)after the treatment of cepharanthine;The expression levels of apoptotic proteins PARP,cleaved-caspase-3 and cleaved-caspase-9 were significantly increased with the increasing concentration of the drug;the flow cytometry results showed that AsPC-1 cells had early and late apoptosis after the treatment of cepharanthine.Conclusion CEP can inhibit the proliferation of pancreatic cancer cells through apoptosis mechanism.
