The Role of PELP1 in Liver Cancer and its Molecular Mechanism

目的探讨脯氨酸-谷氨酸-亮氨酸富集蛋白1(PELP1)参与肝癌发生发展的驱动作用及机制。方法通过使用Oncomine数据库进行数据挖掘分析PELP1在肝癌细胞株或组织中的表达状态,Western blot予以验证。设计并合成3对siRNA沉默肝癌中PELP1的表达,采用Western blot检测转染成功与否。使用成功转染PELP1-siRNA后的细胞株HepG2进行细胞活力实验、平板克隆形成实验、细胞划痕实验、Transwell小室侵袭和迁移实验。Western blot实验检测敲除PELP1后STAT3信号通路表达水平的变化。结果PELP1在肝癌中呈现高表达,通过siRNA沉默PELP1后显著抑制了肝癌细胞的生长活力、侵袭和迁移能力,降低了其恶性程度。Western blot显示将肝癌中PELP1沉默后,降低了磷酸化STAT3的表达。结论PELP1可能参与肝癌的发生发展,具有作为肝癌治疗靶点的潜在价值,参与调控STAT3的激活,从而促进肝癌细胞的生长。

Objective To explore the driving effect and mechanism of proline glutamate leucine enriched protein 1(PELP1)in the occurrence and development of liver cancer.Methods The expression of pelp1 in hepatoma cell lines or tissues was analyzed by data mining using Oncomine database,which was verified by Western blot.Three pairs of siRNAs were designed and synthesized to silence the expression of PELP1 in hepatocellular carcinoma,and then the success of transfection was detected by Western blot.Afterwards,the cell line HepG2 successfully transfected with PELP1-siRNA was used for cell viability experiments,plate clone formation experiments,cell scratch experiments,Transwell cell invasion and migration experiments.The expression level of STAT3 signaling pathway after PELP1 knocked out was detected by Western blot.Results PELP1 was highly expressed in liver cancer.Silencing PELP1 by siRNA significantly inhibited the growth viability,invasion and migration ability of liver cancer cells,and reduced the degree of malignancy.Western blot showed that silencing PELP1 reduced the expression of phosphorylated STAT3 in liver cancer.Conclusion PELP1 may be involved in the occurrence and development of liver cancer and has potential value as a therapeutic target for liver cancer.It is involved in regulating the activation of STAT3,so as to promote the growth of liver cancer cells.
