荷叶黄酮对D-半乳糖诱导小鼠肝损伤的改善作用 被引量:6

摘要 以荷叶黄酮为研究对象,研究其药理价值。通过DPPH自由基、ABTS阳离子自由基清除实验考察其体外抗氧化活性,通过动物实验考察其体内抗氧化能力,雄性昆明小鼠40只平均随机分成4组,采用D-半乳糖诱导建立小鼠肝损伤模型,低、高剂量组小鼠分别口服250、500 mg/(kg·d)荷叶黄酮2~6周。苏木精-伊红染色法切片染色分析肝脏病理变化;测定其肝脏组织中丙二醛(malondialdehyde,MDA)、谷胱甘肽(glutathione GSH)、NO、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)、超氧化物歧化酶(superoxide dismutase,SOD)含量;利用酶联免疫法检测细胞因子IL-6、IL-10、TNF-α、IL-1β的浓度;运用HPLC对荷叶黄酮的种类及其含量进行测定。结果表明,荷叶黄酮具有良好的清除DPPH自由基、ABTS阳离子自由基的能力,能提高小鼠机体组织SOD、GSH、GSH-Px等活性,降低MDA和NO含量,能抑制TNF-α、IL-6和IL-1β的表达,促进IL-10的表达。HPLC分析表明,在荷叶中检测到芦丁、异槲皮苷、紫云英苷、槲皮素4种生物活性成分。 The flavonoids extracted from the lotus leaf were used as the research object.The antioxidant activity of flavonoids was investigated by DPPH and ABTS cation ion free radical scavenging experiments in vitro,and the high antioxidant activity of these extracts was positively correlated with the dose.Forty male Kunming mice were randomly divided into normal group,model group,low dose group and high dose group.The liver injury model was induced by D-galactose,and the model group,low dose group and high dose group were intraperitoneally injected with D-galactose 120 mg/(kg·d).Different doses of lotus leaf flavonoids were used for intervention.Mice in the low-dose and high-dose group were given 250 and 500 mg/(kg·d)lotus leaf flavonoids per day,respectively.The contents of malondialdehyde(MDA),glutathione(GSH),NO,glutathione peroxidase(GSH-Px)and superoxide dismutase(SOD)were determined by using kit colorimetry.The concentrations of cytokines IL-6,IL-10,TNF-αand IL-1βwere determined by ELISA.The organ index and pathological changes were observed.In addition,the variety and content of flavonoids extracted from lotus leaf were determined by HPLC.The results showed that flavonoids had a good scavenging ability of DPPH and ABTS cation ion free radicals,and increased SOD,GSH and GSH-Px content,reduced MDA,NO content significantly.Lotus flavone could significantly improve the liver cell oxidative damage induced by D-galactose in mice.In addition,four bioactive components,rutin,isoquercitrin,astragalin and quercetin,could be detected from lotus leaf by HPLC analysis.In a word,flavonoids extracted from lotus leaf have the good anti-oxidation ability,its pharmacological value can be further studied.
