miR-223-3p在口腔鳞癌细胞增殖、迁移和侵袭中的作用研究

The role of miR-223-3p in the proliferation,migration and invasion of oral squamous cell carcinoma cells

目的:探讨miR-223-3p通过调控NLRP3抑制口腔鳞癌细胞增殖、迁移和侵袭的作用。方法:体外培养NOK细胞和TcaG8113细胞,检测细胞中miR-223-3p、NLRP3和Caspase-1 mRNA表达;将阴性对照(NC)和miR-223-3p特异性mimics转染到TcaG8113细胞中,48 h后检测细胞增殖率、细胞凋亡率、迁移能力和侵袭能力的变化,同时检测转染后TcaG8113细胞中miR-223-3p、NLRP3、Caspase-1、IL-18和IL-1β表达。结果:与NOK细胞相比,TcaG8113细胞中miR-223-3p mRNA表达降低,NLRP3和Caspase-1 mRNA表达增加(P<0.05);转染miR-223-3p mimics后,与NC对照组比较,miR-223-3p mimics组细胞凋亡率和miR-223-3p增加,增殖率、迁移细胞数、侵袭细胞数、NLRP3、Caspase-1、IL-18和IL-1β降低(P<0.05)。结论:miR-223-3p在OSCC细胞中低表达,对OSCC细胞增殖、迁移和侵袭起促进作用,可能通过靶向NLRP3来发挥作用,有可能成为OSCC新治疗的靶点。

Objective:To investigate the effect of miR-223-3 p inhibiting the proliferation,migration and invasion of oral squamous cell carcinoma cells by regulating NLRP3.Methods:NOK cells and TcaG8113 cells were cultured in vitro and the expression of miR-223-3 p,NLRP3 and Caspase-1 mRNA in the cells were detected.Negative control(NC)and miR-223-3 p specific mimics were transfected into TcaG8113 cells,and the cell proliferation rate,cell apoptosis rate,migration ability and invasion abilitywas detected after 48 hours.The expression of miR-223-3 p,NLRP3,Caspase-1,IL-18 and IL-1β in TcaG8113 cells after transfection were also detected.Results:Compared with NOK cells,the expression of miR-223-3p mRNA in TcaG8113 cells was decreased,and the expression of NLRP3 and Caspase-1 mRNA increased(P<0.05).After transfection of miR-223-3p mimics,compared with the NC control group,the apoptosis rate and miR-223-3p were increased in the miR-223-3p mimics group,and the proliferation rate,the number of migrating cells,the number of invading cells,NLRP3,Caspase-1.IL-18 and IL-1β decreased(P<0.05).Conclusion:The low expression of miR-223-3p in OSCC cells promotes the proliferation,migration and invasion of OSCC cells.miR-223-3p may act by targeting NLRP3,and may become a target for new treatments for OSCC.
