Paenibacillus sp.SSG-1来源一种新颖木聚糖酶的克隆、表达、纯化和酶学性质研究 被引量:2

摘要 从土壤类芽孢杆菌Paenibacillus sp.SSG-1中克隆得到木聚糖酶基因xyn30,构建表达载体后,重组蛋白成功表达。通过金属亲和层析对xyn30编码的木聚糖酶进行纯化,并对其酶学性质进行研究。结果表明:以榉木木聚糖为底物,重组木聚糖酶xyn30的最适温度为60℃,在40~70℃范围内,相对酶活力可达75%以上;最适pH为6.0,pH 5.0~9.0范围内,相对酶活力能保持在60%以上;重组木聚糖酶的动力学常数Km值为1.688 mg/mL,Vmax值为3.458μmol/min·mL。
