康复新对去卵巢大鼠学习记忆能力和基底前脑胆碱能神经元的影响 被引量:1

摘要 目的探讨康复新对去卵巢大鼠的学习记忆能力的影响及其可能机制。方法3月龄健康雌性SD大鼠36只,随机分成4组,即假手术组、去卵巢对照组、康复新低剂量组和康复新高剂量组,分别进行干预。8周后用Y迷宫检测大鼠的空间学习记忆能力,用免疫组化染色检测大鼠基底前脑不同区域胆碱能神经元的数目。结果康复新高剂量组大鼠"学会"所需总训练次数、训练错误次数及记忆检测时错误反应次数与去卵巢对照组比较明显减少(P<0.05),与假手术组相比差异无统计学意义(P>0.05);康复新液高、低剂量组大鼠基底前脑内侧隔核(MS)、斜角带核垂直支(vDB)和水平支(hDB)的Ch AT阳性神经元数量增多;高剂量组基底前脑各区的Ch AT阳性神经元数明显高于去卵巢对照组(P<0.05),与假手术组相比差异无统计学意义(P>0.05);而且康复新高剂量组大鼠vDB和h DB的Ch AT阳性神经元数明显高于康复新低剂量组(P<0.05)。结论康复新液可显著改善去卵巢大鼠学习记忆能力,其作用可能是通过增加基底前脑胆碱能神经元的功能实现的;康复新液有望成为防治雌激素撤退引起的神经功能障碍的药物。 Objective To explore the effect of Kangfuxin on learning and memory ability of ovariectomized rats and its possible mechanism.Methods Thirty-six healthy female SD rats aged 3 months were randomly divided into 4 groups,whic are sham operation group,ovariectomized control group,Kangfuxin low-dose group and Kangfuxin high-dose group.Intervention was carried out respectively.Eight weeks later,the spatial learning and memory ability of rats was detected by Ymaze,and the number of cholinergic neurons in different regions of the basal forebrain of rats was detected by immunohistochemistry.Results The total training number,training error number for"learning"and error number for memory test of the Kangfuixn high dose group were significantly lower than the ovariectomized control group(P<0.05),but there was no significant difference compared with the sham operation group(P>0.05);the number of ChAT positive neurons in the medical septum(MS),vertical limb with diagonal band(vDB)and horizontal limb(hDB)of the basal forebrain was increased in the Kangfuxin high and low dose groups.The number of ChAT positive neurons in each region of the basal forebrain in the highdose group was significantly higher than that in the ovariectomized control group(P<0.05),but with no significant difference compared with the sham group(P>0.05);Moreover,the number of ChAT positive neurons in vDB and hDB of rats in the Kangfuixn high-dose group was significantly higher than that in the Kangfuixn low-dose group(P<0.05).Conclusion Kangfuxin solution can significantly improve the learning and memory ability of ovariectomized rats,which may be mediated by increasing the function of cholinergic neurons in basos forebrian.Kangfuxin solution is expected to be a drug for the prevention and treatment of neurological dysfunction caused by estrogen withdrawal.
