核桃JrGL3基因的生物信息学及转录表达分析

摘要 bHLH(basic/Helix-Loop-Helix)转录因子在植物的生长发育及对外界环境胁迫应答中起着非常重要的调控作用,为了分析bHLH家族成员在核桃生长发育过程中作用,采用生物信息学工具对JrGL3结构进行研究,并分析其在不同组织中及低温胁迫下的转录表达。结果表明,JrGL3 cDNA全长为4982 bp,开放阅读框的长度为1914 bp,共编码637个氨基酸;JrGL3蛋白存在典型的bHLH保守结构域,定位于细胞核、无信号肽、无跨膜结构稳定亲水蛋白;通过比对确定了拟南芥中GL3的同源蛋白,并对其进行蛋白互作预测分析,其表明JrGL3与花青素的积累与合成相关;同源分析表明JrGL3与阿月浑子和欧洲栓皮栎的蛋白亲缘关系较近;与梅和橡胶树等植物的蛋白亲缘关系较远。通过低温胁迫“辽宁8号”和“清香”核桃叶片,48 h后转录组表达分析,JrGL3基因呈现显著上升趋势,且在“清香”不同组织转录组分析JrGL3基因在“清香”雌花中表达量最高。 The bHLH(basic/Helix-Loop-Helix)transcription factors played an important regulatory role in plant growth and development and response to environmental stress.In order to analyze the role of bHLH family members in the growth and development of walnut,in this study,bioinformatics tools were used to analyze the structure of the JrGL3 and its transcriptional expression in different tissues and under low temperature stress.The results showed that the full length of the JrGL3 cDNA was 4982 bp,and the length of open reading frame was 1914 bp,encoding 637 amino acids.The JrGL3 protein had a typical bHLH conserved domain,located in the nucleus,no signal peptide,no transmembrane stable hydrophilic protein.The GL3 homologous proteins in Arabidopsis thaliana were identified by comparison,and the protein interaction analysis showed that the JrGL3 was related to the accumulation and synthesis of anthocyanins.Homologous analysis showed that the JrGL3 was closely related to Pistacia vera and Quercus suber.It was distantly related to plants such as Prunus mume and Hevea brasiliensis.After 48 hours of low temperature stress,the expression of the JrGL3 gene in the leaves of‘Liaoning 8’and‘Qingxiang’walnut showed a significant increasing trend,and the expression of the JrGL3 gene in the female flowers of‘Qingxiang’was the highest according to the analysis of the transcriptome of different tissues of‘Qingxiang’.
