Study on the antidepressant effect of Cornus officinalis Sieb. et Zucc.

摘要 目的观测山茱萸抗抑郁作用。方法常规回流提取法制备山茱萸水提物,采用高效液相色谱法(HPLC)测定指标性成分马钱苷含量,在此基础上,将山茱萸水提物连续灌胃给予雄性小鼠14 d,通过开场实验、悬尾实验、强迫游泳实验等行为学测试方法观测山茱萸水提物的抗抑郁作用。结果山茱萸水提物的中、高剂量(150、300 mg/kg)以及阳性药盐酸氟西汀给药14 d后,均可显著降低小鼠在悬尾实验和强迫游泳实验测试的不动时间,而山茱萸低剂量(75 mg/kg)对此无显著影响,且各组之间在开场实验测试的水平运动得分和垂直运动得分相比均无明显差异。结论山茱萸水提物呈现出良好的抗抑郁作用,且具有明显的量效关系。 Objective To observe the antidepressant effect of Cornus officinalis Sieb. et Zucc.. Methods The aqueous extract of C. officinalis was prepared by conventional reflux extraction method, and the content of loganin, an index component, was determined by high performance liquid chromatography. On this basis, the aqueous extract of C. officinalis was continuously administered to male mice by gavage for 14 days. Behavioral testing methods such as tail suspension and forced swimming were used to observe the antidepressant effects of aqueous extract of C. officinalis. Results The medium and high doses(150, 300 mg/kg) of C. officinalis water extract and the positive drug fluoxetine hydrochloride all significantly reduced the immobility time of mice in the tail suspension test and the forced swimming test after 14 days. However, the low dose of C. officinalis water extract(75 mg/kg) had no significant effect on this, and there was no significant difference between the horizontal and vertical movement scores in the opening field test. Conclusion The water extract of C. officinalis shows a good antidepressant effect, and has a significant dose-effect relationship.
