地骨皮炮制前后地骨皮乙素含量变化研究 被引量:5

摘要 目的建立地骨皮中地骨皮乙素含量测定方法,并比较炮制前后地骨皮乙素含量变化。方法采用高效液相色谱法测定地骨皮乙素的含量,色谱条件为Agilent ZORBAX SB-Aq(4.6 mm×250 mm, 5μm),甲醇-0.2%三氟乙酸梯度洗脱,流速1.0 mL/min,检测波长280 nm,柱温40℃。结果地骨皮乙素在24.304 0μg/mL~1.215 2 mg/mL范围内线性关系良好,r^(2)=1.000 0(n=6),精密度试验RSD为0.07%,重复性试验平均含量为3.687 0%,RSD为1.50%,稳定性试验RSD为0.13%,平均回收率为101.53%,RSD为1.73%。结论地骨皮乙素含量随着淘洗次数的增加而显著降低,不同干燥方法对地骨皮乙素含量影响较大,地骨皮乙素含量在一定范围内随着干燥温度的升高而升高。所建立的含量测定方法稳定准确,重现性好,为地骨皮药材和饮片质量控制和评价奠定基础。 Objective To establish a method for the determination of Kukoamine B in Lycii Cortex and to compare the changes of Kukoamine B in Lycii Cortex before and after processing. Methods Agilent ZORBAX SB-Aq(4.6×250 mm, 5 μm), methanol-0.2 % trifluoroacetic acid gradient elution, flow rate 1.0 mL/min, detection wavelength 280 nm, column temperature 40 ℃ was used to determine the content of Kukoamine B. Results In the range of 24.304 0 μg/mL~1.215 2 mg/mL, the linear relationship was good, r^(2)=1.000 0(n=6), the RSD of precision test was 0.07 %, the average content of repetitive test was 3.687 0 %, RSD was 1.50%, the RSD of stability test was 0.13%, the average recovery was 101.53 %, RSD was 1.73 %. Conclusion With the increase of washing times, the Kukoamine B in Lycii Cortex decreased significantly. Different drying methods have a great effect on the Kukoamine B in Lycii Cortex, and the Kukoamine B in Lycii Cortex increased with the increase of drying temperature in a certain range. The established method is stable, accurate and reproducible, which lays a foundation for the quality control and evaluation of Lycii Cortex and decoction pieces.
