摘要
目的探讨慢性阻塞性肺疾病急性加重期(AECOPD)患者平面细胞极性(Wnt/PCP)信号通路中Ras homolog gene family member A(RhoA)蛋白表达与中医证型的相关性,尝试为AECOPD中医证型诊断提供一个客观标准。方法收集痰浊壅肺证、痰热郁肺证、痰蒙神窍证、肺肾气虚证、阳虚水泛证5个证型的AECOPD患者各20例。健康组15例。所有入选患者空腹抽取静脉血,采用Enzyme Linked Immunosorbent Assay(ELISA)检测五个组别血清中RhoA蛋白量,并探讨其相关性。结果与健康组比较,AECOPD患者5个证型的RhoA蛋白量明显增高,具有显著统计学意义(P<0.05);五个中医证型血清中RhoA蛋白量比较:痰浊壅肺组<痰热郁肺组<痰蒙神窍组<肺肾气虚组<阳虚水泛组。RhoA蛋白表达量与AECOPD五组中医证型之间存在直线线性相关关系,且呈正性相关。结论 AECOPD患者Wnt/PCP信号通路中RhoA蛋白量在五个中医证型中存在显著差异,能够给AECOPD中医证型诊断提供一个客观标准,并揭示其疾病严重程度。
Objective To investigate the correlation between the expression of Ras Homolog Gene family Member A(RhoA) protein in the planar cell polarity(Wnt/PCP) signaling pathway in patients with acute exacerbation of chronic obstructive pulmonary disease(AECOPD) and TCM syndrome type, and to attempt to provide an objective standard for the diagnosis of TCM syndrome type of AECOPD.Methods AECOPD patients with five syndromes, namely, turbid-phlegm syndrome, hot-phlegm-stagnation syndrome, Shen-qi syndrome of phlegm and kidney deficiency and Water-flooding syndrome of Yang deficiency, were collected in 20 cases each. The healthy group included 15 patients. Enzyme Linked Immunosorbent Assay(ELISA) was used to analyze the protein content of RhoA in serum of all the five groups.Results Compared with the healthy group, RhoA protein levels of the 5 syndrome types of AECOPD patients were significantly increased(P<0.05). Comparison of RhoA protein amount in serum of five TCM syndromes: The group with phlegm-turbidity obstruction lung < the group with phlegm-heat stagnation lung < the group with phlegm-Meng Shenqi < the group with lung and kidney Qi deficiency < the group with Yang deficiency and water flood. RhoA protein expression was linearly and linearly correlated with TCM syndromes in AECOPD five groups, and the correlation was positive.Conclusion The protein relative expression of RhoA there are differences in the five groups of TCM syndrome type which can provide a objective diagnostic criteria for acute exacerbation of AECOPD TCM syndrome during acute exacerbation period and reveal their disease severity in the Wnt/PCP signaling pathway.
