电针对颅脑损伤大鼠脑组织中促凋亡蛋白Bad、Bak、Bax的影响 被引量:4

Effect of electroacupuncture on proapoptotic proteins Bad, Bak, and Bax in the brain tissue of TBI rats.

摘要 目的探索电针治疗对颅脑损伤(TBI)大鼠损伤脑组织中诱导细胞凋亡蛋白Bad、Bak、Bax的影响。方法将100只SPF级雄性SD大鼠随机分为空白组(10只)、假手术组(30只)、模型组(30只)、电针组(30只)。采用改良Feeney自由落体法制备TBI大鼠模型,于造模后24h介入电针治疗(取"百会、水沟、内关、曲池、足三里、涌泉"穴,1次/天,共14天),分别于电针干预后3天、7天、14天取材,用免疫荧光法检测损伤脑组织中Bad、Bak、Bax蛋白的表达量。结果电针干预后各时间点,假手术组大鼠损伤脑组织中Bad、Bak、Bax的表达与空白组间相比P>0.05,模型组的表达量均高于假手术组(P<0.05);在干预后7天、14天相应时间点,电针组大鼠损伤脑组织中Bad、Bak、Bax的表达量低于模型组(P<0.05)。结论电针干预可下调TBI大鼠模型损伤脑组织中促凋亡蛋白Bad、Bak、Bax的表达。 Objective To explore the effect of electroacupuncture(EA) on apoptosis proteins Bad, Bak and Bax in the injured brain tissue of rats with craniocerebral injury(TBI). Methods 100 SPF male SD rats were randomly divided into blank group(10 rats), sham operation group(30 rats), model group(30 rats) and electroacupuncture group(30 rats). The TBI rat model was prepared by the modified Feeney free-fall method, and electroacupuncture treatment was intervened 24 hours after modeling( "Baihui, Shuigou, Neiguan, Quchi, Zusanli, and Yongquan" points were taken once a day for 14 days), and samples were taken at 3 days, 7 days, and 14 days after electroacupuncture intervention to detect the expression levels of Bad, Bak, and Bax proteins in the injured brain tissue by immunofluorescence. Results At each time point after electroacupuncture intervention, the expression levels of Bad, Bak and Bax in the injured brain tissue of rats in the sham operation group were significantly higher than those in the blank group(P > 0.05), and the expression levels of Bad, Bak and Bax in the injured brain tissue of rats in the electroacupuncture group were lower than those in the model group at the corresponding time points of 7 days and 14 days after intervention(P < 0.05). Conclusion Electroacupuncture can down-regulate the expression of pro-apoptotic proteins Bad, Bak and Bax in the injured brain tissue of TBI rat model.
