长链非编码RNA LINC00963对垂体腺瘤细胞恶性表型的影响

摘要 目的探究长链非编码RNA LINC00963(LncRNA)对垂体腺瘤细胞恶性表型的影响。方法取大鼠垂体腺瘤细胞GH3,将其随机分为空白组、第一转染组和第二转染组。第一和第二转染组分别转染阴性对照片段和LINC00963小干扰RNA(siRNA)干扰序列,空白组细胞不做任何处理。以Trannswell法检测垂体腺瘤细胞GH3侵袭能力,以划痕实验检测垂体腺瘤细胞GH3迁移能力,以四甲基偶氮唑蓝(MTT)法检测垂体腺瘤细胞GH3的增殖抑制率,以原位末端标记(TUNEL)法检测垂体腺瘤细胞GH3凋亡情况。结果空白组、第一转染组和第二转染组侵袭数分别为(93.37±8.86),(91.04±8.71)和(68.71±6.65)个;迁移率分别为(69.27±6.67)%,(63.35±5.94)%和(41.37±3.92)%;增殖抑制率分别为(6.12±0.47)%,(6.43±0.51)%和(19.23±1.78)%;凋亡率分别为(5.14±0.47)%,(3.05±0.29)%和(17.28±1.65)%。第一转染组与空白组相比较,细胞侵袭数、迁移率、增殖率和凋亡率差异均无统计学意义(P>0.05);第二转染组与第一转染组相比较,细胞侵袭数、迁移率、增殖率和凋亡率差异均有统计学意义(P<0.05)。结论长链非编码RNA LINC00963可促进垂体腺瘤细胞侵袭、迁移和增殖能力,并减少细胞凋亡。 Objective To investigate the effects of long non-coding RNA LINC00963 on GH3 invasion,migration,proliferation and apoptosis in pituitary adenoma cells.Methods Rat pituitary adenoma cells GH3 were randomly divided into blank group,Transfection-1 group and transfection-2 group.Transfection-1 group and transfection-2 group were transfected with NC fragments and LINC00963-siRNA transfected with interfering sequences,respectively,and blank group cells were left untreated.Trannswell method was used to detect GH3 invasion of pituitary adenoma cells,scratch assay was utilized to detect GH3 migration of pituitary adenoma cells,proliferation inhibition rate of GH3 in pituitary adenoma cells by methyl thiazolyl tetrazolium(MTT)method,and TUNEL staining were conducted to detect GH3 apoptosis in pituitary adenoma cells.Results The number of invasion cells in the blank group,transfection-1 group and transfection-2 group were 93.37±8.86,91.04±8.71,68.71±6.65,respectively;the migration rates were(69.27±6.67)%,(63.35±5.94)%,(41.37±3.92)%;the proliferation inhibition rates were(6.12±0.47)%,(6.43±0.51)%,(19.23±1.78)%,respectively;the apoptosis rates were(5.14±0.47)%,(3.05±0.29)%,(17.28±1.65)%,respectively.There were no significant differences in the number of cell invasion,migration rate,proliferation rate and apoptosis rate between the transfection-1 group and the blank group(P>0.05);compared with the first transfection group,the invasion number,migration rate,proliferation rate and apoptosis rate of cells in the transfection-2 group were statistically significant(P<0.05).Conclusion Long non-coding RNA LINC00963 promoted pituitary adenoma cell invasion,migration and proliferation,and also reduced apoptosis in pituitary adenoma cells.
