摘要
目的探讨环状RNA_0005692(circ_0005692)在前列腺癌中的表达及功能。方法获取57例前列腺癌患者肿瘤组织和癌旁组织,采用实时荧光定量聚合酶链反应(qRT-PCR)检测肿瘤组织和癌旁组织中circ_0005692的表达。将人前列腺癌细胞系22RV1培养后随机分为4组:pcDNA3.1-circ_0005692组、NC组、si-circ_0005692组和si-NC组,分别转染circ_0005692过表达质粒、空白质粒、circ_0005692小干扰RNA和无意义小干扰RNA。用细胞计数试剂盒-8(CCK-8)实验检测各组细胞的存活率;用TUNEL实验检测各组细胞凋亡率;用Transwell实验检测各组细胞侵袭能力;用Western blot实验检测细胞增殖相关蛋白Ki67和上皮间质转化相关蛋白N-钙黏蛋白(N-cadherin)、波形蛋白(Vimentin)和E-钙黏蛋白(E-cadherin)的相对表达量。结果circ_0005692在前列腺癌患者肿瘤组织中的表达水平(0.51±0.12)显著低于癌旁组织(1.02±0.45),差异有统计学意义(P<0.001)。NC组、pcDNA3.1-circ_0005692、si-NC组和si-circ_0005692组的细胞存活率分别为(100.17±10.15)%,(75.65±6.25)%,(85.98±10.13)%和(134.25±14.57)%;这4组的细胞凋亡率为(8.35±0.93)%,(18.48±3.43)%,(9.65±1.23)%和(5.12±0.67)%;这4组的细胞侵袭数目分别为110.22±9.76,57.41±8.55,90.25±10.35和169.26±17.54;这4组的Ki67蛋白相对表达量为0.24±0.04,0.09±0.01,0.30±0.02,1.02±0.13。上述指标,NC组和pcDNA3.1-circ_0005692组比较,si-NC组和si-circ_0005692组比较,差异均有统计学意义(均P<0.05)。结论circ_0005692在前列腺癌中显著下调,可抑制前列腺癌细胞增殖、侵袭和上皮间质转化。
Objective To explore the expression and function of circular RNA_0005692(circ_0005692)in prostate cancer.Methods Tumor and paracancerous tissues were obtained from 57 patients with prostate cancer.The expression of circ_0005692 in tumor tissues and paracancerous tissues of patients with prostate cancer was detected by real-time fluorescence quantitative polymerase chain reaction(qRT-PCR).Prostate cancer cell lines 22RV1 were cultured and randomly divided into 4 groups:pcDNA3.1-circ_0005692 group,NC group,si-circ_0005692 group and si-NC group,which were transfected with circ_0005692 overexpressed plasmid,blank plasmid,circ 0005692 small interfering RNA and nonsense small interfering RNA.The cell survival rate of each group was detected by cell counting kit-8(CCK-8)assay.The cell apoptosis rate of each group was detected by TUNEL assay.Transwell assay was used to detect the invasion ability of cells in each group.The relative expression of cell proliferation-related protein Ki67 and epithelial-mesenchymal transition related proteins N-cadherin,Vimentin and E-cadherin were detected by Western blot assay.Results The expression level of circ_0005692 in tumor tissues(0.51±0.12)of prostate cancer patients was significantly lower than that in paracancerous tissues(1.02±0.45),and the difference was significant(P<0.05).The cell survival rate of the NC group,the pcDNA3.1-circ_0100196 group,the si-NC group and the si-circ_0005692 group was(100.17±10.15)%,(75.65±6.25)%,(85.98±10.13)%and(134.25±14.57)%;the cell apoptosis rate was(8.35±0.93)%,(18.48±3.43)%,(9.65±1.23)%and(5.12±0.67)%,respectively;the number of invaded cells was 110.22±9.76,57.41±8.55,90.25±10.35 and 169.26±17.54;the Ki67 protein expression was 0.24±0.04,0.09±0.01,0.30±0.02,1.02±0.13.Compared between the NC group and pcDNA3.1-circ_0005692 group,as well as the si-NC group and si-circ_0005692 group,the difference was significant(P<0.05).Conclusion Circ_0005692 was significantly down-regulated in prostate cancer and it can inhibit the proliferation,invasion and epithelial-mesenchymal transition of prostate cancer cells.
