山姜素对冠心病大鼠心功能、血脂和血管内皮功能的影响 被引量:9

Effect of alpinia on heart function,blood lipid and vascular endothelial function in rats with coronary heart disease

摘要 目的研究山姜素对冠心病大鼠心功能、血脂和血管内皮功能的影响及机制。方法将SD大鼠分成对照组、模型组(冠心病模型)、阳性对照组(辛伐他汀治疗)和低、中、高剂量实验组(冠心病模型,10,80,160 mg·kg^(-1)山姜素)。用彩色多普勒超声诊断仪检测心功能指标左心室舒张末期容积(LVEDV),用全自动生化分析仪测定血清中三酰甘油(TG),用Griess法检测血清中一氧化氮(NO)含量,用酶联免疫吸附(ELISA)法检测血清中内皮素-1(ET-1)、肿瘤坏死因子-α(TNF-α)含量,用蛋白质印迹(Western blot)法检测MEK/胞外信号调节激酶(ERK)信号相关蛋白p-MEK/MEK表达。结果对照组、模型组、阳性对照组和低、中、高剂量实验组大鼠LVEDV分别为(4.15±0.23),(6.21±0.57),(5.19±0.60),(5.60±0.45),(5.04±0.61),(4.39±0.31)mL,血清中TG含量为(0.77±0.07),(1.61±0.09),(1.16±0.15),(1.46±0.11),(1.24±0.12),(0.99±0.13)mmol·L^(-1),NO含量分别为(40.38±3.37),(27.43±1.36),(37.93±2.25),(30.95±2.34),(34.32±2.58),(39.11±0.22)μmol·L^(-1),ET-1含量分别为(4.12±0.38),(12.03±1.38),(7.39±0.48),(9.85±0.08),(7.16±0.47),(6.34±0.80)ng·L^(-1),TNF-α含量分别为(16.74±1.99),(39.56±4.06),(22.67±2.87),(31.55±4.29),(26.64±2.36),(21.08±1.81)pg·mL^(-1),胸主动脉组织中p-MEK/MEK表达量分别为0.29±0.03,1.00±0.06,0.37±0.05,0.76±0.06,0.57±0.07,0.41±0.03。上述指标,模型组与对照组比较,差异均有统计学意义(均P<0.05);阳性对照组和低、中、高剂量实验组与模型组比较,差异均有统计学意义(均P<0.05);中、高剂量实验组比较,低、中实验组比较,差异均有统计学意义(均P<0.05)。结论山姜素可能通过抑制MEK/ERK信号通路减轻冠心病大鼠炎症,进而改善血管内皮功能、血脂和心功能。 Objective To investigate the effect and mechanism of alpinia on heart function,blood lipid and vascular endothelial function in rats with coronary heart disease.Methods SD rats were divided into control group,model group(coronary heart disease model),positive control group(simvastatin treatment)and low,medium,high dose experimental groups(coronary heart disease model,alpinia 40,80,160 mg·mL^(-1)treatment).Color doppler ultrasound diagnostic instrument was used to detect cardiac function indicators left ventricular end-diastolic volume(LVEDV),automatic biochemical analyzer was used to determine serum triglyceride(TG),Griess method was used to detect serum nitric oxide(NO)content,enzyme linked immunosorbent assay(ELISA)method was used to detect serum endothelin-1(ET-1),tumor necrosis factor-α(TNF-α)content,Western blot method was used to detect MEK/extracellular signal regulate the expression of extracellular regulated protein kinases(ERK)signal-related proteins p-MEK/MEK.Results The LVEDV of control group,model group,positive control group and low,medium,high dose experimental groups were(4.15±0.23),(6.21±0.57),(5.19±0.60),(5.60±0.45),(5.04±0.61),(4.39±0.31)mL,the content of TG in the serum were(0.77±0.07),(1.61±0.09),(1.16±0.15),(1.46±0.11),(1.24±0.12),(0.99±0.13)mmol·L^(-1),the contents of NO were(40.38±3.37),(27.43±1.36),(37.93±2.25),(30.95±2.34),(34.32±2.58),(39.11±0.22)μmol·L^(-1),ET-1 content were(4.12±0.38),(12.03±1.38),(7.39±0.48),(9.85±0.08),(7.16±0.47),(6.34±0.80)ng·L^(-1),TNF-αcontent were(16.74±1.99),(39.56±4.06),(22.67±2.87),(31.55±4.29),(26.64±2.36),(21.08±1.81)pg·mL^(-1),the expression of p-MEK/MEK in rat thoracic aorta tissue were 0.29±0.03,1.00±0.06,0.37±0.05,0.76±0.06,0.57±0.07,0.41±0.03.The differences of the above indicators between model group and control group were statistically significant(all P<0.05);differences between positive control group,low,medium,high dose experimental groups and model group were statistically significant(all P<0.05);the difference between low and medium dose experimental groups,between medium and high dose experimental groups were all statistically significant(all P<0.05).Conclusion Alpinia may reduce inflammation in rats with coronary heart disease by inhibiting the MEK/ERK signaling pathway,thereby improving vascular endothelial function,blood lipids and heart function.
