原花青素B2对脓毒症大鼠心肌损伤的作用 被引量:8

Effect of procyanidin B2 on myocardial injury in septic rats

摘要 目的研究原花青素B2对脓毒症大鼠心肌损伤的作用。方法将SD大鼠分为空白组、模型组、对照组(乌司他丁)和低(50 mg·kg^(-1)原花青素B2)、中(100 mg·kg^(-1)原花青素B2)、高剂量实验组(200 mg·kg^(-1)原花青素B2)。以彩色多普勒超声诊断仪检测心功能指标左心室舒张末期内径(LVEDD)、左心室收缩末期内径(LVESD)、短轴缩短分数(FS)和射血分数(EF),检测心肌组织中超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量,以酶联免疫吸附(ELISA)法检测心肌组织中炎症因子水平,以蛋白质印迹法检测B细胞淋巴瘤/白血病-2(Bcl-2)、Bcl-2相关X蛋白(Bax)、剪切的胱天蛋白酶3(cleaved caspase-3)、腺苷酸蛋白活化激酶α(AMPKα)、磷酸化的AMPKα(p-AMPKα)、核因子-κB p65(NF-κBp65)蛋白表达。结果空白组、模型组、对照组和低、中、高剂量实验组LVEDD分别为(5.60±0.52),(7.99±0.55),(5.82±0.27),(7.06±0.56),(6.51±0.40),(5.93±0.39)mm,LVESD为(2.95±0.30),(7.32±0.54),(4.44±0.56),(6.29±0.27),(5.25±0.40),(4.63±0.46)mm,FS为(46.05±5.58)%,(26.12±3.99)%,(37.29±4.12)%,(32.55±3.84)%,(38.38±2.28)%,(43.38±3.43)%,EF为(76.72±5.69)%,(40.60±5.57)%,(64.60±4.73)%,(49.02±2.76)%,(56.86±3.44)%,(67.27±3.90)%,上述指标,模型组与空白组比较,差异均有统计学意义(均P<0.05);对照组和低、中、高剂量实验组与模型组比较,差异均有统计学意义(均P<0.05);中、低剂量实验组比较,高、中剂量实验组比较,差异均有统计学意义(均P<0.05)。结论原花青素B2可改善脓毒症大鼠心功能,改善心肌组织炎症和氧化应激,机制可能与AMPK/NF-κB信号通路有关。 Objective To study the effects of proanthocyanidin B2 on myocardial injury in septic rats.Methods SD rats were divided into blank group,model group,control group (ulinastatin) and low (50mg·kg;procyanidin B2),medium (100 mg·kg;procyanidin B2)and high dose experimental (200 mg·kg;procyanidin B2) groups.The left ventricular end-diastolic diameter (LVEDD),left ventricular end-systolic diameter (LVESD),fractional shortening (FS) and ejection fraction (EF) were detected by color Doppler ultrasonography.The activity of superoxide dismutase (SOD) and malondialdehyde(MDA) in myocardium were detected.The levels of inflammatory factors in myocardium were detected by enzyme-linked immunosorbent assay(ELISA).The expression of B cell lymphoma/lewkmia-2 (Bcl-2),Bcl-2 associated X protein (Bax),cleaved caspase-3,adenosine protein-activated kinaseα(AMPKα),phosphorylated AMPKα(P-AMPKα),and nuclear factor-κB p65 (NF-κBp65) proteins were detected by Western blot.Results The LVEDD in blank group,model group,control group and low,medium and high dose experimental groups were (5.60±0.52),(7.99±0.55),(5.82±0.27),(7.06±0.56),(6.51±0.40),(5.93±0.39) mm,LVESD were (2.95±0.30),(7.32±0.54),(4.44±0.56),(6.29±0.27),(5.25±0.40),(4.63±0.46) mm,FS were (46.05±5.58)%,(26.12±3.99)%,(37.29±4.12)%,(32.55±3.84)%,(38.38±2.28)%,(43.38±3.43)%,EF were(76.72±5.69)%,(40.60±5.57)%,(64.60±4.73)%,(49.02±2.76)%,(56.86±3.44)%,(67.27±3.90)%.There were statistically significant differences between model group and blank group (all P<0.05).There were statistically significant differences between low,medium,high dose experimental groups,control group and model group (all P<0.05).There were statistically significant differences between low and medium dose experimental groups,and between high and medium dose experimental groups (all P<0.05).ConclusionProanthocyanidin B2 can improve cardiac function,improve myocardial inflammation and oxidative stress.The mechanism may be related to AMPK/NF-κB signal pathway.
