九藤逐瘀方对血瘀型输卵管阻塞性不孕症大鼠血清Bcl-2、EGFR及sICAM-1影响及病理组织学观察

Effect and Pathohistologlogical Observation of Jiuteng Zhuyu Decoction on the Serum Level of Bcl-2,EGFR and sICAM-1 in the Blood Stasis Model Rats with Tubal Obstruction Infertility

目的观察九藤逐瘀方不同给药途径对血瘀型输卵管阻塞性不孕症大鼠血清B细胞淋巴瘤-2(Bcl-2)、肾小球滤过率(EGFR)及人可溶性细胸间粘附分子(sICAM-1)的影响。方法将80只Wistar雌性大鼠,按随机数字表法均分为8组。除空白对照组外,各组大鼠均于造模后15 d开始给药,至九藤逐瘀方药干预30 d后,用酶联免疫吸附法(ELISA)检测大鼠血清Bcl-2、EGFR及sICAM-1的水平,并于各组大鼠输卵管取材,进行常规苏木素伊红(HE)染色,光镜观察。结果九藤逐瘀方能降低输卵管阻塞性不孕症大鼠血清EGFR及sICAM-1的水平,升高血清中Bcl-2的水平。用九藤逐瘀方干预的6组大鼠输卵管结构通过肉眼观察均比模型组有不同程度的改善,尤其以九藤逐瘀方灌肠高剂组明显,已接近正常表现。从光镜下输卵管组织病理学改变情况来看,九藤逐瘀方灌肠组与灌胃组都较模型组有不同程度的恢复,以九藤逐瘀方灌肠高剂组表现最好,基本接近正常。结论九藤逐瘀方药对血瘀型输卵管炎性不孕症大鼠具有很好的治疗作用。

Objective To observe the effect of Jiuteng Zhuyu decoction on the serum level of Bcl-2,EGFR and sICAM-1 in the blood stasis model rats with tubal obstruction.Methods 80 female Wistar rats were selected and randomly divided into eight groups.We made the model with Jiuteng Zhuyu decoction after the model has been taken 15 days in addition to the normal control group from the fallopian tube of each group of rats.ELISA method was adopted to determine the level of Bcl-2,EGFR and sICAM-1,and then we made HE staining and made optical observation with continuously doing Jiuteng Zhuyu decoction 30 days.Results Jiuteng Zhuyu decoction can decrease the serum level of EGFR and sICAM-1,increase the serum level of Bcl-2 in the tubal obstruction infertility rat models.Compared with the rats in the model group,the oviduct structure in rats of six groups intervened with vine herbs turned better to different degree by raw eyes,especially in high dose of Jiuteng Zhuyu decoction clysis group whose oviduct structure was generally normal.Observation under optical microscopes showed that histopathological changes in oviduct of six intervention groups were differently improved,and the effect was the best in high doses of Jiuteng Zhuyu decoction clysis group,which was basically normal.Conclusion Jiuteng Zhuyu decoction treated fallopian tube obstructive infertility rats had achieved good results.
