摘要
基于铂包金纳米粒子(Au@Pt NPs),建立一种高灵敏度、高选择性的啶虫脒间接竞争ELISA检测方法。方法中的Au@Pt NPs能催化氧化3, 3’, 5, 5’-四甲基联苯胺产生蓝色氧化产物以实现定量检测。结果表明该检测体系的最佳条件为:用不含甲醇的PBS缓冲液稀释啶虫脒标品,用50 mmol/L pH 8.0的PBS缓冲液稀释特异性抗体及Au@Pt NPs-IgG,加入显色液孵育20 min。在最优条件下建立的间接竞争酶联免疫检测方法,其对啶虫脒的检测线性范围为0.57~35.70 ng/mL,最低检出限达0.167 ng/mL。
A high-sensitivity and high-selectivity indirect competitive enzyme-linked immunoassay(ELISA) immunoassay for the detection of acetamiprid was established based on Au@Pt nanoparticles. In this method, Au@Pt nanoparticles could catalyze the oxidation of TMB and produce the blue oxidation product to realize quantitative detection. The optimal condition of this method was using PBS buffer without methanol to dilute acetamiprid, 50 mmol/L PBS buffer with pH 8.0 to dilute the specific antibody and Au@Pt NPs-IgG and incubating 20 min. Under this condition, the linear range was from 0.57 ng/mL to 35.70 ng/mL, and the minimum detection limit was 0.167 ng/mL.
