基于5-HT研究逍遥丸对肝郁雌性大鼠性激素及卵巢功能的影响

Effects of Xiaoyao Pills on sex hormones and ovarian function in female rats with liver depression based on 5-HT

目的观察逍遥丸调节五羟色胺(5-HT)对大鼠性激素和卵巢功能的影响。方法采用束缚法建立雌性大鼠肝郁模型,糖水摄取实验及旷场行为学实验评估情志状况判断建模效果,阴道涂片监测动情周期,HE染色观察卵泡个数,ELISA检测血清性激素、抗缪勒管激素(AMH)水平高效液相检测下丘脑5-羟色胺(5-HT)、5-HT自身受体(5-HT1A)。逍遥丸灌胃3周治疗后,检测上述指标。结果与空白组比较,模型组大鼠糖水消耗量及糖水偏爱程度均明显降低,旷场移动总距离及直立次数减少动情周期紊乱,卵泡数量减少,下丘脑5-HT降低,血清性激素血清雌二醇(E2)、黄体生成素(LH)、及AMH水平下降,卵泡刺激素(FSH)升高,均具有统计学差异(P<0.05或P<0.01);与模型组比较,逍遥丸各组糖水消耗量及糖水偏爱程度均升高(P<0.05或P<0.01),动情周期紊乱有所改善,卵泡数量增多(P<0.05),下丘脑5-HT、5-HT1A升高(P<0.05),血清FSH降低,E2、AMH、LH水平均升高(P<0.05或P<0.01);与西药组比较,中药中剂量组糖水消耗量及糖水偏爱程度均升高动情周期紊乱有所改善,直立次数增加,卵泡数量增多,下丘脑5-HT、5-HT1A升高血清FSH降低,E2、AMH、LH水平均升高。疗效依次为:中药中剂量组> 西药组>中药高剂量组。结论逍遥丸通过调节5-HT影响性激素改善卵巢功能。

Objective To observe the effects of Xiaoyao Pills on sex hormones and ovarian function in rats.Methods The liver depression model of female rats was established by binding method.The emotional status was evaluated by sugar water intake experiment and open field behavior experiment,and the modeling effect was judged.The estrous cycle was monitored by vaginal smear,the number of follicles was observed by HE staining,the levels of serum sex hormones and anti-mullerian hormone(AMH) were detected by ELISA,and the hypothalamic serotonin(5-HT) and 5-HT autoreceptor(5-HT1A) were detected by HPLC.The above indexes were detected after intragastric administration of Xiaoyao Pills for 3 weeks.Results Compared with blank group,the sugar water consumption and sugar water preference of model group were significantly reduced,the total distance of open field movement and the number of upright times were reduced,the estrous cycle was disordered,the number of follicles was reduced,the 5-HT of hypothalamus was reduced,the levels of serum sex hormones,serum estradiol E_(2),luteinizing hormone(LH),AMH and follicle stimulating hormone(FSH) were increased(P<0.05 or P<0.01);compared with model group,the sugar water consumption and sugar water preference of Xiaoyao Pills group increased(P<0.05 or P<0.01),the disorder of estrous cycle improved,the number of follicles increased(P<0.05),5-HT and 5-HT1A in hypothalamus increased(P<0.05),serum FSH decreased,and the levels of E_(2),AMH and LH increased(P<0.05 or P<0.01);compared with western medicine group,the sugar water consumption and sugar water preference in middle dose group of TCM increased,the disorder of estrous cycle was improved,the number of upright times increased,the number of follicles increased,the hypothalamic 5-HT and 5-HT1A increased,the serum FSH decreased,and the levels of E_(2),AMH and LH increased.The order of curative effect was:middle dose group of TCM> western medicine group> high dose group of TCM.Conclusion Xiaoyao Pills can regulate 5-HT,affect sex hormones and improve ovarian function.
